Corydalis yanhusuo W. T. Wang is herb, perenial. It belongs to Corydalis of Fumarioideae of Papaveraceae. We use it’s dry tuber in medicine. Corydalis is recognized worldwide as a good painkiller and C. yanhusuo plays an important role in Corydalis, which has great economic value and is an important medicinal plant resource. In recent years, affected by environmental damage and excessive excavation, C. yanhusuo wild resources under serious threat, which can easily result in depletion of germplasm. Therefore, we use the materials from East China, Central China, North China, South China, North-west of China as materials to conduct experiment in C. yanhusuo.In this study 19 populations included 380 individuals from five regions above was used to analyze the genetic diversity. In the last, we develope a corresponding protection strategy according to the experiment.1. This study initially used Chenggu, Hanzhong, C. yanhusuo of fresh leaves and tubers as materials, Comparing CTAB method, improved CTAB method, SDS method with improved SDS method to the DNA extraction, we optimize the method of DNA extraction. The results showed: improved CTAB method was suitable for the extraction of Corydalis leaf DNA; SDS extraction method was suitable for corydalis tuber DNA.2. The development of microsatellite primers. Extracted the whole genome DNA from C. yanhusuo, libraries are constructed. Used second-generation molecule sequencing technology to assemble and screen, microsatellite sequences were designed 36 pairs of SSR primers. C. yanhusuo was used as the as materials to do the experiment about polyacrylamide gel electrophoresis uerder the method of improved CTAB. At last, 12 pairs of high polymorphism, good stability primers were screened out, laying the foundation for subsequent genetic analysis Corydalis diversity.3. Using dried leaves of C. yanhusuo from 19 populations included 380 individuals and improved CTAB method to polyacrylamide gel electrophoresis, using 12 pairs of SSR primers were screened, we analysis the genetic diversity C. yanhusuo. Result showed that, in the population level, genetic diversity of C. yanhusuo is low.( HE = 0.3979, h = 0.3879, I = 0.6132). At the species level, abundant genetic variation of C. yanhusuo is abundant( HE = 0.6351, h = 0.6343, I = 1.1926). C. yanhusuo genetic differentiation among populations more significant: FST = 0.3883. AMOVE analysis showed that 47.28% of genetic variation between populations located in which 25.94% of the variation between the three geographic units occur. NM = 0.3938, gene exchange described fumarate populations lower; Mantel Test analysis showed that the genetic distance between populations fumarate their respective geographical distance showed a significant positive correlation(r = 0.326, p = 0.01), show that geographic isolation plays an important role in the current genetic structure of C. yanhusuo populations. Principal component analysis and cluster analysis UPGMA are C. yanhusuo from 19 populations were divided into three groups. C. yanhusuo genetic structure is mainly based on cross-breeding system, clonal growth, geographic isolation and gene flow. Based on the genetic structure of C. yanhusuo, we proposed to in situ conservation-based conservation strategies.
