In order to enhance the level of differential diagnosis of tumor disease in pet clinical, the application research carry on canine tumor disease in ultrasound imaging and molecular biology level. The key to carry out ultrasound elasticity imaging technology 、P53 gene in the differential diagnosis of research on canine mass disease. And to further explore the application value of the above two kinds of new diagnostic technique in the pet clinical. Based on the pet dog as the research object in this paper, expect that for pet mass disease clinical differential diagnosis to provide a certain basis and theoretical guidance.The Experiment of ultrasound imaging studies selected clinical conform to the requirements of the pet dog tumors 18 cases, the results of the cases were statistically analyzed, eighteen canine cases of tumors in ultrasound diagnosis accordance with the principle of analysis ultrasound images and the method of collecting samples at the same time. Using the method of tissue pathological analysis of eighteen canine mass cases. Experimental base breeding female beagle six health standard as a negative control, collect clinical eight cases of female pet dog malignant breast tumor samples as positive group. Semi-quantitative determination of analysis P53 gene in the mammary gland organization to make use of ordinary PCR, using the real-time quantitative(real time Q-PCR) method at the RNA level quantitative determination of P53 gene expression.The pet dog happening of the mass disease in the clinical on age, gender, variety and parts with certain rules to follow. In the system of ultrasound elasticity imaging, the application of ultrasound elastography technology in the diagnosis of mass of benign and malignant for accuracy was 94.4%(17/18). Single according to the elastic score, strain ratio, the elastic area ratio of canine mass and combined application of diagnosis of benign versus malignant masses benign and malignancy, results of the comprehensive compared with of strain ratio(88.9%) no significant difference(P=0.06,P>0.05). PCR determination results indicate, the experimental group compared with the negative control group, gel electrophoresis banding imaging gene brightness and width was significantly higher, real-time quantitative PCR results showed, in the negative control group, result CT value of P53 gene and GAPDH gene are greater than 12.5, in the experimental group, result CT value of P53 gene and GAPDH gene are less than 10.0, analysis of the result shows that P53 gene expression in malignant breast tumor tissue volume and the amount of P53 gene expression in normal breast tissue were significantly difference, especially the number 3 significant difference(P<0.05) among them, the others extremely significant difference( P<0.01) among them.The result shows that ultrasound elasticity imaging in distinguishing good dog tumors compared with the traditional ultrasonic technology has obvious specificity and accuracy. The technology can be used as a clinical pet dogs in the future mass is an important identification method of benign and malignant. P53 gene expression level can be used as a clinical monitoring of pet dogs breast mass disease an important molecular index in the future.
