Objective A wild Cricetulus migratorius is a wild rodent animal living in central asia.It belongs toCricetulus, Cricetinae, circetidae, Rodentia. Its habitat is mainly distributed in Plain Desert, alpine meadow.Most studies show that the Cricetulus migratorius have advantage of Yersinia pestis isolated and Aspectsof hydatid disease model, has a good application prospect. The Research Center for Experimental Animalsof Xinjiang reseach Cricetulus migratorius as experimental animal for many years. Morphologicalobservation and gene identification to the strain of flagellate which was isolated from Cricetulusmigratorius in the Research Center for Experimental Animals of Xinjiang; To set up an experimental modelof flagellate infection in mice, Screening of deworming drugs and to study its anthelmintic effect..Methods The cultivation in vitro were established, the flagellate was cultured in1640medium in vitro. Theflagellate was observed microscopically direct smear and conventional Giemsa staining, Total DNAisolated from XinJiang Cricetulus migratorius was extracted and16s rRNA was amplified by RCR,then sequenced. Furthermore the homology was compared and the phylogenetic tree was developed usingMEGA5.22. The flagellate which cultured in vitro infected with BALB/c mice to established experimentalmodel, To observe the effects of Metronidazole, tinidazole, Albendazole Oral Emulsion, FurazolidoneTablets and Compound Dihydroartemisinin Tablets on flagellate. Results We found that the flagellatesparasitic in the grey hamster ileal cecum by dissecting microscope, also a small insect was found in ileum;Morphological observation Preliminary showed that the flagellate isolate was Tritrichomonas muris. The16S rRNA gene sequence of the XinJiang Cricetulus migratorius isolate shared highly homology with thatof other Tritrichomonas. Phylogenetic tree analysis indicated that the16S rRNA gene of the XinJiangCricetulus migratorius isolate was classified into a subgroup with T. muris16S rRNA(U85966.1), but wasdistant relative with other related tritrichomonas. Experimental infections were established throughintragastric administration with105Tritrichomonas muris cultured in1640medium polypide vaccinatedinto BALB/c mice which were6to8weeks old on3consecutive days.Metronidazole and furazolidonegroups have obviously adverse reaction in the experiment;The count of Tritrichomonas muris trophozoitesshowed that metronidazole and tinidazole groups of all BALB/c mice were0, have significant effects.Compound dihydroartemisinin group, Tritrichomonas muris trophozoites of female (4.54±0.53×104/mg,p=0.000) and male Cricetulus migratorius (4.48±0.51×104/mg, p=0.000)was reduced significantly. Thealbendazole group and furazolidone group counting results compared with the solvent control group haveno significant difference. Conclusion The flagellate isolate in XinJiang Cricetulus migratorius wassuccessfully identified to be T. muris by both the morphological observation and the16S rRNA geneanalysis. Pharmacodynamic experiments show that the tinidazole inhibited and killed Tritrichomonas muristrophozoites thoroughly and it has the short course of treatment, less adverse drug reactions;Metronidazolehas obvious killing effect,but it has more adverse drug reaction; Albendazole, furazolidone treatmentshowed no obvious killing effect and the adverse reactions of drugs significantly; Dihydroartemisinin hascertain curative effect, adverse drug reaction wass not obvious.
