| In this paper, the Cerasus tianschanica Pojark in wild-fruit forests were used as test materials whichfrom the Da Xigou valleys,Huocheng County,Xinjiang,China.The Cerasus tianschanica Pojark stems,tissue culture technology to explore from the sterile system, initial culture, proliferation culture, to thewhole process of rooting; the delicate branches and cutting,and study the different growth-regulatinghormone in the delicate branches and the deck take root in effect; Studied on removing seed shell,differentsoaking time and different hormones treatment of Cerasus tianschanica Pojark germination and radiclegrowth. Main results are as follows:1,Cerasus tianschanica Pojark first generation culture,rooted in material sterilization to70%alcohol30s+HgCl212min best; MS in media and has begun the burgeoning rate higher than B5media,which ignitedrate reached93.4%of Early generation develop the best recipe to MS+1.5mg/L6-BA+0.1mg/LIBA+30g/Lsugar+6g/L agar.2,Cerasus tianschanica Pojark subculture of plantlets best proliferation medium for MS+6-BA1.60mg/L+IBA0.10mg/L+GA30.10mg/L, proliferation coefficient as high as8.07, the average numberof bud had9.33and stem length had10.53mm while subculture of plantlets arrived35d. In the stratification andseed soaking time phase at the same time, the seed of radicle growth was higher than that of shell, thelongest is28.59mm.Cerasus tianschanica Pojark shelled seed germination rate decreased gradually withthe increased of GA3concentration, explain GA3has certain inhibition on germination rate of the Cerasustianschanica Pojark, Inhibition with laminated seed treatments decreased gradually with the extension oftime and time24h was higher than12h on germination rate of seed soaking, and increase with the extensionof stratification time.Cerasus tianschanica Pojark to low temperature stratification above70d seed coat,different concentration of GA3and24h radicle growth was higher than seed soaked with12h; Radicle up to53.40mm. |
