Most of Gesneriaceae plants are of high ornamental value and medicinal value. It is the distribution and diversity center of China for the subfamily Cyrtandroideae. In recent years, most of the Gesneriaceae plants with very narrow distribution range, the special habitat and fragmented distribution are in danger because of the participation of human activities and the destruction of the habitat.Deinocheilos jiangxiensis belongs to the genera of Deinocheilos in Trib.Didymocarpeae of Cyrtandroideae, established in1986by Academician Wang wencai. It’s endemic genera of China, which has only three distribution points throughout our country, less and endangering. The researches of biological characteristics and breeding are the basis of the study of biodiversity protection. This paper would study for Deinocheilos jiangxiensis from these two aspects, establishing the tissue culture system of rapid propagation and providing the related data supports for the species of the wild regression test. The experimental results are as follows:(1)The community of Deinocheilos jiangxiensis in sample survey distributed on the cliff of the crook by the roa, population focused, fewer. The structure of the associated plants tree layered the average height of4m, whose dominant species contained Torreya jackii, Acer pauciflorum et.The shrub layered the average height of1.5m, whose dominant species contained some plants of Ericaceae and Rosaceae. The herb layered the average height of0.4m, whose dominant species contained mosses and ferns et.Mosses and fems became typical associated species in the community which enjoy the cool and damp environment. The community sat on the edge of the scenic highway, affected by human activities.(2)The Deinocheilos jiangxiensis populations of flowering in greenhouse was18days in October (August to October in wild habitat), single flower bloomed4-5days, pale purple corolla. From the point of flower structure:corolla funnel-shaped cylinder;4ventricles, placentas parietal; capsule linear.2crack; seeds small, brown, short cylindrical. The matured time difference of arrival with anther and stigma, and the presence of foreign media to pollinate, became the important factors of seed setting rate.(3)Using orthogonal experiment design method to discuss different parts of explants,culture conditions, sucrose concentration, kind and concentration of exogenous hormones on the induction of somatic embryogenesis and the influence of strong seedling roots, we got the best somatic embryo differentiation medium:1/2MS+6BA2.0mg-L-1+IBA0.02+2,4-D0.5+far from petiole segments explant+dark condition training;the best callus induction differentiation media:MS+6-BA2.0+TDZ0.02+far from petiole segments explant+dark condition training;the best combination for rooting medium:l/8MS+NAA0.2+IBA0.2+sucrose40g-L-1.Statistics indicate that low concentration of cytokinin could induce the formation of callus and somatic embryos, and high concentration of cytokinin decreased the ability of callus and somatic embryogenesis, with the browning phenomenon also increasing.Low concentration of auxin could effectively promote the callus induction, high concentrations of auxin could effectively promote the happening of the embryo.(4)The seedling transplanting survival rate of Deinocheilos jiangxiensis is high, even the import peat and mixing matrix could reach above90%, the vermiculite only56%, which had the lowest survival rate of the experiment.In addition to the important factors such as temperature, humidity, the transplanting time and fine management also have direct impacts on the transplanting survival rate of the tissue culture plants.(5) The somatic embryogenesis of Deinocheilos jiangxiensis observed with the method of paraffin section included such several stages:single cell proembryo, multicellular proembryo, globular embryo, heart embryo, torpedo embryo and cotyledon embryo. In addition, the secondary embryogenesis inductions were observed during the period of globular embryo, and it existed isolation phenomenon during the period of cotyledon embryo.
