| The study using the immature inflorescence, immature embryo, mature embryo of a new Russian wildrye strian(Psathyrostachys juncea) as tissue culture materials. Those material were cultured on MS culture medium. Study on effects of period of material on tissue culture, the treatment before culture, exogenous and other nutriments on callus induction of the new Russian wildrye Strian. Study on the feasible subculture medium, as a result of the poor quality of the callus, also investigating in the differentiation and roots production. The results are concluded as follows:(1)Immature inflorescence culture system : The optimum length of immature inflorescence is 1~2cm, the optimum medium of callus inducing is Y4:MS + 2,4-D(4.0mg/L),the frequency of induction is up to 100%; the optimum medium of subculture is J4:MS + 2,4-D(1.0 mg/L)+ 6-BA(0.5mg/L),the frequency of embryogenic callus is 46.15%; the optimum medium of differentiation is F9:MS + NAA(0.5mg/L)+ 6-BA(1.0mg/L), the frequency of differentiation is 94.59%.(2)Immature embryo culture system:The optimum immature embryo age is about 14d, the optimum treatment time with low temperature of 4℃is 1d, the optimum medium of callus induction is YA1:MS + 6-BA(0.2mg/L)+ 2,4-D(2.0mg/L)+ ABA(0.3 mg/L), the callus was improved by added 0.3mg/L ABA, though the frequency of induction a bit decreased; the optimum medium of subculture is J1:MS + 2,4-D(0.5mg/L),the frequency of embryogenic callus is 66.34%; the optimum medium of differentiation is F7:MS + NAA(0.3mg/L)+ 6-BA(4.0mg/L),the frequency of differentiation is 81.03%.(3)Mature embryo culture system:The optimum seed soaking time is 48h; the optimum medium of callus induction is Y7C:MS + 6-BA(0.2mg/L)+ 2,4-D(1.0mg/L)+ CH(500mg/L),the induction frequency is 75%; the optimum medium of subculture is J2:MS + 2,4-D(0.5 mg/L)+ 6-BA(0.5mg/L), the frequency of embryogenic callus is 37.33%; the optimum medium of differentiation is F7:MS + NAA(0.3mg/L)+ 6-BA(4.0mg/L),the frequency of differentiation is 85.29%.(4)The optimum medium of roots production is SG2:1/2MS + NAA (0.5 mg/L), the frequency of roots production is 100%. |
