| In this paper using seeds, leaf of test-tube ptandets and stem as explants, throughobserving induction callus and the producing process of somatic embrgos, analysingthe hormone condition and formation path on dedifferentiation and redifferentiationmedium. In order to offer theory and practice bases through tissue culture tointermediate propagation, secondary metabolism clone variation and seed selection.The results were as follows:1. The induction ratios of stem of test-tube plandet was higher than leaf; theincreased fresh weight and the increased dry weight of the callus which was inductedby leaf of test-tube plandet both higher than the stem of test-tube plandet.2.In different porportion of plant hormone, the medium with NAA and 6-BAhave the best effect on the induction ratio,growing and quality of callus.3. When we inducted callus using seeds as explant, the induction ratios washigher with different component of NAA and dense 6-BA.; When we used leafs asexplant to induct callus.; the low component of NAA and most component of 6-BAshowed higher induction ratios. When the condition was on the contrary, it showedlower induction ratios.4. Effect of differentiation ratios in induction medium and differentiation medium,some differentiation medium had excellent effect in different induction medium(E1),and some differentiation medium (J)and induction medium had no connection, buthad high differentiation ratio.5. There were organogenesis and somatic embryogenesis on molThogenesis ofThe Rhodiola Sachalinensisi A. Bor, it was same to other dicotyledon, they allexperience four previous stages of global embryo, heart-shaped embryo, torpedoembryo, and cotyledon embryo.6. The cotyledon embryo happened inside or cover of callus, When we used seedsas explant most of cotyledon embryo produced from the cover of callas. When weused tube-test plandet as explant the most produced inside callus. |
