“Ge QI” Chinese herb compound screened out by animal pharmacy lab,Shanghai Veterinary Research Institute,Chinese Academy of Agricultural Science, is a novel Chinese traditional medicine formula.This formula is based on the theory of the Chinese veterinary dialectical treats,which has good antidiarrheal effect.To investigate the preventive and therapeutic effect of this formula, preclinical pharmacy tests were conducted,including the study about the effect of the formula on the diarrhea mouses infected with E.coli;the study about the establishment of piglets diarrhea models infected with E.coli; the study about the effect of the formula on the diarrhea piglets infected with E.coli;the study on the bacteriostatic test of kinds of traditional Chinese herbs and foumulas,which provide tests data for the approval of “Ge QI ”Chinese herb compound. 1 Study about the effect of the formula on diarrhea mouses infected with E.coliThe pathological model of diarrhea mouse infected with E.coli was pre-established, then group 0.3mL/10 g,group 0.2mL/10 g,group 0.1mL/10 g,model control group and blank control group were designed, next each dose group was orally administrated 7days continuously and observed the effect of this formula on the mouses.The results showed that the average weight gain,hematology index,spleen and thymus index in dose group were basically the same with the blank control group,whereas significantly different( P < 0.05) from the model control group. Duodenum histopathological examination showed that there were no obvious pathological changes in each dose group, while in model control group the duodenum mucosa became swollen and thicker.We inferred that the formula could prevent and cure the mouse diarrhea. 2 Study about the pathological model of diarrhea piglets infected with E.coliGroup5mL/10 Kg 、 group2.5mL/10 Kg 、 group1mL/10 Kg and blank control group were designed.Each dose group was orally infected with 3×109CFU/mL E. coli bacterial suspension 3days continuously by the weight.The results showed that the average weight gain, bacteriological examination, hematology index and electrolyte index in high and medium dose were significantly different from blank control group(P<0.05).It was inferred that the piglets diarrhea models could be established by orally infected with 3×109CFU/mL E. coli bacterial suspension 3days continuously in a 5mL/10 Kg dose. 3 Study about the effect of the formula on diarrhea piglets infected with E.coliThe pathological models of diarrhea piglets infected with E.coli were pre-established, then group 10mL/10 Kg,group 5mL/10 Kg,group 2.5mL/10 Kg,model control group and blank control group were designed, next each dose group was orally administrated 7days continuously and observed the effect of this formula on the piglets.The results showed that the clinical symptoms,the average weight gain,white blood cell(WBC),A/G, the electrolyte index,such as Na,K in each dose group were basically the same as the blank control group,while significantly different from the model control group( P < 0.05). Duodenum histopathological examination showed that there were no obvious pathological changes in each dose group,while in model control group the intestinal epithelial and glands mortified and shedded, the inflammatory cells in the mesenchyme of lamina propria increased and were infiltrated, what’s more,the demaged organizational structure could not get restored.We inferred that formula could prevent and cure the piglets diarrhea. 4 The study about bacteriostatic test of several kinds of traditional Chinese herbs and formulas against swine E. coli in vitro.Inhibitory effect of several kinds of Chinese herbal medicine and foumulas were explored for swine Escherichia coli(E.coli)in vitro by using the agar plate diffusion method and trace double dilution method. The diameter of the zone of inhibition and minimum inhibitory concentration(MIC) was determined.The results showed that Coptis chinensis, Scutellaria baicalensis,etc., and the foumula were sensitive for swine E.coli,and have good antibacterial effects in vitro.
