Bioactive Components Investigation And Tissue Culture Of A New Commercial Cultivar Chuanbeichai No.1 In Bupleurum Chinese DC.

Radix Bupleuri is one of the most important medical herbs in China, Japan, Korea and other Asian countries. It is often combined with other drugs to treat colds, fever, asthenic fever, chills etc. Because of the important medicinal value, the demand of Bupleurum has increased dramatically. Therefore, it is urgently to explore Bupleurum varieties with high-yield and high-quality to ensure large area of cultivation for Bupleurum ind meet the market demand. We have investigated the yield, bioactive components and tissue culture of a new commercial cultivar ChuanbeichaiNO.1 in Bupleurum Chinese DC.in present study, by contrast with the commercial varieties of ZhongchaiNO.1 and ChuanhongchaiNO.1 to provide a technical ind theoretical basis for validation and application of ChuanbeichaiNO.1. The major results in present study are as ’ollows:ZhongchaiNO.1 showed the highest dry weight in all investigated environments. The dry weight of root of ChuanbeichaiNO.1 was 10.9% lower than ZhongchaiNO.1 and ChuanhongchaiNO.1 has lowest dry weight. ChuanbeichaiNO.1 showed higher bioactive components content of it than ZhongchaiNO.1 and ChuanhongchaiNO.1. Its content of total saponins, total flavonoids, polysaccharides and Mg are higher than ZhongchaiNO.1 with 3.5%,16.33%,16.37% and 4.73% respectively, and higher than ChuanbeichaiNO.1 with 60.34%, 36.7%,22.55% and 18.21% respectively. The result of component content investigation in different organs of Bupleurum showed that the total saponin, carbohydrates and trace element content in root is higher than which in stems and leaves. And the flavonoids in stems and leaves are higher than that in root of Bupleurum. The content of saponin, polysaccharide and Mg in root is higher than which in stems and leaves of ChuanbeichaiNO.1 with 72.86%,28.01%,33.18% respectively, but he total favonoids in stems and leaves is higher than which in root with 58.45%. The above results showed that ChuanbeichaiNO.1 is an excellent new commercial cultivar. It showed significant advantage in the content of bioactive components. This line can be recommended authorize of new commercial varieties of Bupleurum and application in Sichuan area.The dry weight of Bupleurum root in Qingchuan is the highest, which was higher than which in Wangcang and Anxian with 23.07% and 32.69%. In bioactive components content investigation, the total saponins, total flavonoids, polysaccharides and Mg of Bupleurum in Qingchuan are higher than which in Wangcang with 20.09%,15.11%,10.98%, and 15.54%, and higher than which in Anxian with 60.34%,36.7%,22.55%, and 18.21%. Therefore, Qingchuan is better for Bupleurum cultivation than Wangcang and Anxian.10% NaClO treatment ChuanbeichaiNO.1 30min is the optimal lisinfection conditions of tissue culture, and contamination rate was 6.7%, the survival rate was 86.7%.6-BA, KT, NAA and IAA have positive effect for induction the cluster buds of ChuanbeichaiNO.1 in single factor testing. The optimal concentration of 6-BA, KT, NAA and AA was 1,0.5,0.2 and 1.0 mg·L-1 respectively. The best combination of auxin levels in induction cluster uds of ChuanbeichaiNO.1 was 0.5mg·L-1 6-BA and 0.5mg·L-1 [AA in the synergistic effect test of various factors. The best cluster buds were obtained under that auxin condition, its average number, length, fresh weight, dry weight and chlorophyll content was 11,4.3cm,1015.9mg,97.8mg and 43.6 respectively.NAA, IBA and IAA have positive effect for induction the adventitious roots of ChuanbeichaiNO.1 in single factor testing. The optimal concentration of NAA, IBA and IAA was 0.1,1.0and 1.0 mg·L-1 respectively. The best combination of auxin levels in the induction adventitious roots of ChuanbeichaiNO.1 was 0.3mg·L-1 NAA, 0.1mg·L-1 IBA and 0.1mg·L-1 IAA in the synergistic effect test of various factors. The best adventitious roots were obtained under that auxin condition, its average number, length, fresh weight, dry weight and chlorophyll content was 105,3.2cm,930.5mg,123.4mg and 25.7 respectively.
