Selenium(Se) deficiency induces various kinds of diseases in human and any other animals including poultry, Nowadays, Farmers prefer to use premix diets with energy feed(corn) and / or protein feed( soybean).Not only the different manufacturers, but also the different place of origin, the selenium content is at different levels. Therefore, due to the detection technology in raw materials, resulting in dietary selenium content in absolute or relative deficiency. Dietary selenium deficiency can not only cause myopathy white chickens, exudative diathesis and pancreatic atrophy, but also the immune dysfunction are the main clinical manifestations of selenium deficiency. To make sure the situation of selenium content in premix diets of broilers, we investigated the premix diets in 11 kinds of brands and detected the selenium content of corn and soybean from different places in Heilongjiang province. With Se deficiency in neutrophils of broilers as a model, detect the mRNA expression of selenoproteins, cytokines(IL-1β, IL-1γ, IL-2, IL-4, IL-6, IL-7, IL-10, IL-12β, IFN-γ and TGF-β4), heat shock protein(Hsp90, Hsp70, Hsp60, Hsp40 and Hsp27)and inflammatory cytokines(COX-2, i NOS, TNF-α, NF-κB and PTGES)by using real-time PCR.Western blotting of heat shock protein(Hsp90, Hsp70 and Hsp60) protein content The results showed that:(1) The selenium content of different plants grow in different soils, selenium content of commercially available premixed feed even meet the standards, but if the selenium content of feed energy and protein feed diets still cause selenium deficiency, selenium deficiency caused by disease.(2) Se deficiency could cause the expression of cytokines. The expression of IL-1β, IL-1γ,IL-6 and IL-7 increased, while the expression of IL-2, IL-4, IL-10, IL-12β, IFN-γ, and TGF-β4 decreased, suggesting that Se deficiency change the function of neutrophilsby regulating the expression of cytokines.(3) Se deficiency could influence the expression of Hsps and Hsp90, Hsp70, Hsp60, Hsp40, Hsp27 had an increased trend in neutrophils of broilers.Hsp40 increased obviously among others, indicated that Hsp40 may play a major role in neutrophil immune cell function.(4) The expression of inflammation factor including COX-2, i NOS, TNF-α, NF-κB and PTGES could be influeced by Se deficiency.And the expression of PTGES, i NOS and TNF-α were higher than others, suggesting that Se deficiency is one of the mechanisms in leading the inflammatory injury of neutrophil dysfunction.(5) All the selenoproteins under detected were at downward trend, but the degree of decline was slight different, which may be related to the distribution of the relevant parts of the body.
