Corydalis decumbens(Thunb.)Pers which belongs to the poppy purple pansy generaand mainly distributed in jiangxi, zhengjiang and other plances,is a traditional Chinesemedicinal materials with a long history in our country. In recent years due to marketdemand and environmental destruction,lead to wild resources decreased rapidly, at thesame time artificial cultivation have obtained rapid development. Tubers of Corydalisdecumbens which used to reserve seed for planting in artificial cultivation easily affectedby plant diseases,inset pests and climate,led to a decline in its quality and yields.Therefore,this eexperiment do a tissure culture test with materials which come fromYujiang Jiangxi,to provide theoretical basis for breeding system for Corydalis decumbensgermplasm resources conservation and set up a new type of non-toxic germplasmresources.1. The tuber, buds and petiole of Corydalis decumbens were taken as explants forcallus induction, different concentrations of hormones were used, to explore the conditionsfor tissue culture and rapid generation of C.decumbens. The results showed that1/2MS+6-BA(2mg/L)+NAA(1mg/L)could promoted the induction of tuber callus withbest effect,frequency of callus induction was59.38%;Induced effect was ideal when takenbuds as explants,especially treated with1/2MS+6-BA(3.5mg/L)+NAA(0.1mg/L)andcallus induction was100%, most of the callus were in induced and germinated later.Whenpetiole callus were induced in1/2MS+6-BA(3mg/L)+2,4-D(2mg/L), frequency of callusinduction was13.39%,but callus could not germinated later.2. Bud germination, proliferation and the plantlets were induced from tuber ofCorydalis decumbens using tissue culture based on different concentrations of hormones,tuber cutting parts and different temperatures. The results showed that the best inducedtemperature was16℃for the sprouting of tubers,bud proliferation and growth of thetuber plantlets. The best medium for tuber sprouting was1/2MS+6-BA(2.5mg/L)+NAA(0.1mg/L)+GA3(0.5mg/L); the optimal medium for bud proliferation was1/2MS+6-BA(3.5mg/L)+NAA(0.1mg/L)+GA3(0.5mg/L). The tuber with skin had good resultsregarding bud propagation, in which the average number of induced shoots was3.71. Themultiple sprounts growed vigorously and formed plantlet easily. The1/2MS basic mediumwithout any hormone was the optimal medium for plantlet proliferation.3. Microtuber were induced from tuber of Corydalis decumbens using tissue culturebased on different concentrations of hormones, sucore concentrations, different lightingand different temperatures. The results showed that1/2MS add concentrations of80g/Lsucore,the rate was highestand the Microtuber induction was86.67%,the plants weregrowth better in the later; The microtuber induction was lower in the1/2MS that adddifferent concentrations of hormones, the microtuber were growth poor; The best inducedtemperature was16℃and the light was the12hours a day for the microtuber.
