Study On Culture In Vitro And Microtuber Induction Of Purple Potato "Zuyun No.1"

The technology of plant tissue culture was adopt in this study, the purple potato "Ziyun No.1" was used as the experimental materials. First, the effects of explant sterilization and different cultural temperatures on the bud explant induction were studied. Then the in vitro cuttings were selected as the sterilizated materials and the medium formula and the cultureal condition of buds multiplication, callus induction and differentiation, microtubers induction, plantles rooting were studied. The results showed that:1. To the sterilization of the bud explants of purple potato "Ziyun No.1", the effects of two factors treatments were better than the single factor treatments. The suitable sterilizated combination was0.14%HgCl214min+13%NaClO10min, the contamination rate of buds explants was24.44%and the bud induction rate was68.89%. The suitable culture temperature of bud induction was26℃, the bud contamination rate and induction rate were37.78%and55.56%.2. MS basic medium was better than other basic mediums on the buds multiplication of purple potato "Ziyun No.1", the bud multiplication multiple was4.00.3. IBA was the suitable auxin on purple potato "Ziyun No.1" bud multiplication, and the best hormone combintation for bud multiplication of "Ziyun No.1"was6-BA3.0mg/L+IBA0.3mg/L and the bud multiplication multiple was5.30. Added TDZ0.5mg/L into the bud multiplication medium of purple potato "Ziyun No.1""was advantage to the bud differentiation and multiplication, the bud multiplication multiple was6.38.4. Petiole was the suitable inoculation site of purple potato "Ziyun No.1" on callus induction,2,4-D was more suitable for callus induction than6-BA, the callus showed light yellow granular and the rate of callus induction was100.00%.5. The best bud differentiation medium of "Ziyun No.1" was MS+6-BA2.0mg/L+IBA0.2mg/L+GA33.0mg/L and the differentiation rate of petiole was92.80%.6.6-BA was the absolutely necessary hormone on microtuber induction of purple potato "Ziyun No.1" and the suitable concentration of6-BA was1.0mg/L.7. It could improve the induction effect of "Ziyun No.1" microtuber when added lOmg/L Coumarin,the tuberization rate was64.44%, the average number of tuber formation was1.62number and microtuber weight was0.028g.8. Different concentrations of sugar could affect the formation and development of "Ziyun No.1" microtuber,45g/L sugar concentration could get the best induction effect for microtuber. The tuberization rate, the average number of tuber formation and microtuber weight were78.33%,1.63number and0.052g.9. In the same microtuber induction medium,the induction effect of basal part of stem was better than the middle or the top part.10. The induction effect of the "Ziyun No.1" microtuber was the best when cultural temperature was22℃. The tuberization rate was79.56%, the average number of tuber formation was1.70number and microtuber weight was0.052g.11. The suitable auxin on rooting of purple potato "Ziyun No.1" was IBA, and the rooting effect of the hormone combintration6-BA0.2mg/L+IBA0.5mg/L+NAA0.4mg/L was the best. The rate of root induction was100.00%, the average of root number was4.64number, average of root diameter was3.86mm, average of root length and average plant height was3.47cm and6.57cm.12. The plant growth inhibitor PP333could improve the induction effect of "Ziyun No.1" rooting,the suitable concentration was0.3mg/L. The rate of root induction was100.00%, the average of root number was4.95number, average of root diameter was3.92mm, average of root length was3.47cm and average plant height was6.57cm.
