Eupolyphaga is one of the traditional Chinese herbal medicines that can activate blood and dissolving stasis. It is rich in active ingredients and is widely applied in pharmaceuticals. With the development of medicine and molecular biological technologies, researchers home and abroad have made deeper and deeper research in eupolyphaga. More pharmaceutical values were found as to-be-developed medicine used for various diseases prevention and treatment with promising application.Hyperlipidemia has an increasingly detrimental effect on human’s health, causing high attention of researchers home and abroad. Development of medicine of low toxicity and high efficiency for treatment of hyperlipidemia has been a research focus.In this research, high fat diet was used to obtain the exogenous hyperlipidemia model of rabbit and the rabbit hyperlipidemia were treated with high, medium and low dose of eupolyphaga powder, which aimed at the treatment of eupolyphaga powder to hyperlipidemia, its mechanism and the optimal dosage.1502.5months old healthy female rabbits whose weights were (1.8±0.2) kg were randomly divided into two groups:blank control group (30) and model group (120). The time of model preparation was50days and after model preparation, the model group was divided into4groups:high, medium, low dosage of eupolyphaga powder treatment group and model control group, each of the group has30rabbits. The high, medium and low dosage were0.45g/kg..0.3g/kg、0.15g/kg respectively and the treatment period was30days. The treatment of eupolyphaga powder to hyperlipidemia and its mechanism were analyzed by the determination of serum lipid parameter (TC, TG, LDL, HDL) of hyperlipidemia rabbits and expression quantity of serum lipid metabolism related genes (ApoE, LDL-R) by RT-PCR. The results showed that:1. After model preparation, the serum lipid levels of rabbits in model group were abnormal and the levels of TC, TG, LDL in serum increased significantly compared to blank control group (p<0.05), while HDL level in the serum decreased significantly.2. After30-day treatment of high, medium and low dosage of eupolyphaga powder, the serum level of rabbits in three groups all recovered. TC, TG, LDL level in serum decreased and HDL level increased significantly compared to model control group (p<0.05). The medium dosage was the optimal.3. Determination of the expression quantity of APoE and LDL-R genes by RT-PCR. After model preparation, the expression quantity of ApoE and LDL-R genes in liver of model group rabbits decreased significantly compared to blank control group (p<0.05). After the treatment of eupolyphaga powder, the expression quantity of ApoE and LDL-R genes in liver of high, medium and low dosage group all increased significantly compared to control group (p<0.05). The results meant that eupolyphaga powder could up regulate the expression of serum lipid metabolism related genes and the high, medium dosage had the best effect.Conclusion:eupolyphaga powder could regulate lipid metabolism disorder in hyperlipidemia and up regulate the expression of serum lipid metabolism related genes (ApoE, LDL-R), therefore, it could treat hyperlipidemia and the optimal dosage was medium dosage.
