Eupolyphagais a kind of traditional Chinese medicine, also known as turtle worms, earth element. It is salty and cold, and with little toxin, owned by the liver, it can break bleeding and continue bones. With the development of science and technology, the research of Eupolyphagais increasingly wide, and with the study of medicinal ingredient and mechanism about Eupolyphaga, it is well known by more people, currently it has wide clinical application in the regulation of lipid and anti-clotting.This traditional Chinese medicine is gradually playing a new vitality on the road for human health services with in-depth study of Eupolyphaga.Lyophilization is freeze drying technique, it means to freeze some substance containing large amounts of water into a solid, and sublimate water vapor under vacuum conditions sublimate out of the ice shelves and remain in the material itself in the freeze. Drying at a low temperature, some of the volatile components of the material loss is very small, and the growth of microbial enzymes can not being, so its original character will be able to maintain. Freeze drying technique is Suitable for some chemical products, pharmaceuticals and food drying. Freeze-drying has application prospects in the pharmaceutical industry, it has have profound significance to explore its use in traditional Chinese medicine formulations.Hyperlipidemia refers to the high lipid levels, there is no uniform standard diagnostic methods, hyperlipidemia can directly cause some serious harm to human health and disease, such as atherosclerosis, coronary heart disease, pancreatitis. With the ever-increasing incidence of hyperlipidemia and its complications, the study of its causes and diagnostic methods catch more and more attention for the Chinese and Western scholars. Medical establishment is one of the important methods in animal models of modern medical research, and has played an important role in the study of disease diagnosis and drug action mechanism, more and more research has been applied to a variety of diseases. In recent years, Chinese and Western scholars use a variety of methods and apparatus to product a variety of medical animal model from different ideas and approaches.In which, the animal models of hyperlipidemia for in-depth study of hyperlipidemia pathogenesis and treatment mechanism is currently one of the most studied.This study built a rabbit model of hyperlipidemia with high-fat diet fed by a natural way, and fed different doses of freeze-dried powder after the success of its modeling Eupolyphaga, and explored the regulation of blood lipids of freeze-dried powder powder of Eupolyphaga and to deter mine the optimal dose and effective time,and analyzed spatial and temporal expression pattern of lipid metabolism-related genes in rabbit ApoE gene and LDL-R gene in rabbit liver by using molecular techniques, investigated the molecular mechanisms regulating blood lipids of eupolyphaga. The main findings are as follows:1)When model is finished, the model rabbits abnormal lipid levels, levels of TC, TG and LDL-C in serum were significantly elevated,they were significantly different (P<0.05) compared with the control group, after different doses of freeze-dried powder of Eupolyphaga treatment TC, TG and LDL-C levels decreased significantly, which TC levels significantly different compared with the model group,it mean indicating Eupolyphagafreeze-dried powder powder can regulate blood lipid levels hyperlipidemia rabbits, which is the optimum dose of0.3g/kg, and25days markedly period.2)When model is finished, ApoE and LDL-R gene of the rabbit liver in model group decreased significantly in expression levels, compared with the control group the difference was significant (P<0.05), after ate different doses of freeze-dried powder Eupolyphagatreatment, each dose treatment rabbits liver was significantly increased in ApoE and LDL-R gene expression levels, compared with the model group significantly different (P<0.05), it described Eupolyphagafreeze-dried powder powder can play their ApoE gene by regulating gene expression and LDL-R regulation of blood lipids, which results middle dose and high-dose group was the best.In summary, this study confirms Eupolyphagafreeze-dried powder can regulate lipid hyperlipidemic rabbits efficacy, and optimal dose is0.3g/kg, markedly period is25days, To regulate ApoE gene and LDL-R gene in the liver and expression may be one of the molecular mechanisms regulating blood lipids.
