High fat canines model were selected as research subject to investigate the effects of green tea powder and green tea polyphenols on high fat canines.Thirty-four male Beagles with the same age(13-14 month),close body weight(12.03-13.93 kg)and similar physiological conditions were selected.(1)Low fat group: low fat diet;(2)High fat group: high fat diet;(3)2.00% GTP group: high fat diet + 2.00% green tea powder;(4)0.48% TPs group: high fat diet + 0.48% tea polyphenols;(5)0.96% TPs group: high fat diet + 0.96% of tea polyphenols;(6)1.92% TPs group: high fat diet +1.92 % tea polyphenols.The test period last for 126 days with drinking water regularly.On the last day of the experiment,all canines were took non-anticoagulant blood with empty stomachand which were used to measure serum indexes.And four canines in each group were injected with Lumianning(0.1 ml/kg)and killed to collect the organes.Abdominal aorta was used for the production of paraffin sections to observe the morphological changes.The liver and adipose tissues were collected for electron microscopy to observe the accumulation of lipid droplets in the organs.Experiment results showed that:1.During the experiment,the levels of serum TG,TC,and LDL-C in high fat group were higher than those in low fat group(P<0.01)and the serum HDL-C levels in high fat group were lower than those in low fat group(P<0.05).The concentrations of serum TG,TC and LDL-C showed a decreasing trend and serum HDL-C showed an increasing trend in 2.00% GTP group,0.48%,0.96% and 1.92% TPs groups.2.During the experiment,compared with low fat group,the concentration of high fat group liver GSH-Px and liver NO were decreased significantly(P<0.01 or P<0.05).Compared with high fat group,the concentration of serum MDA decreased significantly(P<0.05),liver T-AOC increased significantly(P<0.01)and liver MDA decreased significantly(P<0.01)in 2.00% GTP group.In 0.96% TPs group,the concentrations of serum GSH-Px,CAT,T-AOC and NO were higher(P<0.05)than that in high fat group.In the 1.92% TPs group,the concentrations of serum SOD,CAT,T-AOC were higher(P<0.05),serum MDA was lower(P<0.05)and the concentration of liver SOD and T-AOC were higher(P<0.01)and liver MDA was lower(P<0.05)than those in high fat group.3.Compared with low fat group,liver IL-1β was higher(P<0.05)in high fat group.Compared with high fat group,the concentrations of serum and liver IL-6 decreased significantly(P<0.05)in 0.48% TPs group.In 0.96% TPs group,the concentrations of serum and liver IL-6 decreased significantly(P<0.05).In 1.92% TPs group,the concentration of serum IL-1β was lower(P<0.05)and the liver IL-1β and IL-6 were decreased significantly(P<0.05).4.Compared with low fat group,spleen index decreased significantly(P<0.05)in high fat group.Compared with high fat group,the concentrations of serum IgA and liver IgG increased significantly(P<0.05)in 2.00% GTP group.The concentration of serum IgG was higher(P<0.05)in 0.96% TPs group,serum IgA,IgG,IgM were higher(P<0.05)and liver IgA,IgG were higher(P<0.05)in 1.92% TPs group than that in high fat group.5.Compared with low fat group,the canine aortic smooth muscle cells were swollen and arranged disorderly in the high fat group.In 2.00% GTP group,0.48%,0.96% and 1.92% TPs groups,aortic smooth muscle cells curved arrangement,aortic wall were slightly thick and endometrial surface were not smooth.The results from transmission electron microscopy showed that there were lipid droplets of different sizes in each of the 5 groups fed with high fat diet and large lipid droplets were found in the canine liver and adipose tissue in high fat group.Lipid droplets were also found in 2.00% GTP group,0.48%,0.96% and 1.92% TPs groups while lipid droplets in these four groups become smaller and decrease.In summary,2.00% GTP,0.48%,0.96% and 1.92% TPs can regulate the balance of lipid metabolism,improve antioxidant capacity,inhibit the production of pro-inflammatory cytokines,increase immunoglobulin content,reduce the accumulation of lipid droplets in the liver and adipose tissue. |
