In this study,Wei and Large White pigs are seleceted as lean and fat type experimental materials to study the contrast between the serum lipid traits,carcass and meat quality traits,selecteing apolipoprotein E(APOE)、 cholesteryl ester transfer protein(CETP)and 3-hydroxy-3-methyl glutaryl coenzyme A reductase(HMGCR)which plays an important role in the blood cholesterol metabolic pathway as a candidate gene.PCR-SSCP Technique was used to detect APOE and CETP and HMGCR section of genetic variation and a comparative analysis of the correlation between liver and longissimus muscle APOE 、 CETP and HMGCR mRNA expression level and pig carcass and meat traits using the GLM procedure of SPSS.In order to screen important functional polymorphism sites as a new molecular genetic marker,to explore with the candidate genes as molecular markers used in the feasibility of pig genetics and breeding,provides a scientific basis for genetic improvement of Anhui local pig breeds blood fat and meat quality traits.The results are as follows:(1)Comparison of blood lipid,carcass and meat quality traits between Wei pig and Large White,TC,TG,HDL-C,LDL-C,backfat thickness,the cholesterol content in muscle,the IMF,the stearic acid content and oleic acid were significantly higher than that of large white(P < 0.01),the content of hexadecylenic acid was higher than large white pigs(P < 0.05),the content of linoleic acid was significantly lower than large white(P <0.01).The relationship of Wei pig’s blood lipid,carcass and meat quality traits showed that there was a significant positive correlation between the blood lipid traits and between lipid traits and carcass and meat quality traits exist different degree of a significant positive correlation,negative correlation with linoleic acid.Large White’s blood lipid,carcass and meat quality traits showed that there was also a significant positive correlation,between lipid traits and carcass and meat quality traits,and negative correlation with linoleic acid.(2)APOE,CETP and HMGCR mRNA have different levels of expression in different tissues of Wei pig,the highest expression in the liver,and a high degree of expression in longissimus and backfat.(3)There was no mutation in APOE gene in three populations;A G→T mutation in intron 1 at 853 bp and a T→C mutation in intron 1 at 858 bp were found in CETP gene in three populations;five mutaions were found in HMGCR gene,an A→C mutation in intron1 at 6380 bp,an A→G mutation in intron 4 at 8409 bp,a T→Cmutation in exon 11 at14295 bp which caused amino acid change from methionine to threonine,a C→T mutationin intron 18 at 20060 bp and a G→A mutation in exon 19 at 20529 bp.By digestion also found that A → G mutation site(c.807A> G)in the presence of exon 9,is synonymous mutations.The contents of TC,HDL-C and LDL-C were significantly different,and the difference of TG content was not significant in different genotypes of individuals at the two mutation sites of CETP gene in Wei pig and large white;The contents of TC,HDL-C and LDL-C were significantly different,and the difference of TG content was not significant in different genotypes of individuals at the three exron and one intron mutation sites of HMGCR gene in Wei pig and large white;the expression of HMGCR gene in two different genotypes individuals also showed significant differences.(4)In liver,the expression of the APOE mRNA and CETP mRNA had a significant negative association with backfat thickness and stearate of the Wei pigs,and the expression of HMGCR mRNA had a significant positive correlation with backfat thickness,but has opposite relationship with stearic acid;the expression of APOE,CETP and HMGCR mRNA in the muscle of Wei pig,had a significant positive correlation with backfat thickness,muscle cholesterol,intramuscular fat,hexadecenoic acid and oleic acid,which was significant negative correlated with stearic acid and linoleic acid.
