Objective:The study of the subject, one observed in obese rats during endurance exercise, exogenous leptin lipid metabolism enzyme activities, explore the exogenous leptin on obesity movement Lipid Metabolismand discussed the role of leptin to reduce fat; Second, is observed in obese rats during endurance exercise, exogenous leptin in rats antioxidant enzymes and the change of substance, so as to explore leptin on obesitymouse anti-oxidation ability, and discuss the impact of anti-fatigue movement of leptin in obese rats. Third, the correlation analysis of the indicators of lipid metabolism and antioxidant explore the lipid metabolism and antioxidant capacity contact.Methods:In this study, it was the obese rats for the study, the obese model succeed by feeding high-fat diet around half time modeling success, the obese rats is400.78±7.73g. The rats were divided into obese group (O group, n=8), without any intervention natural feeding; obese and exercise group (OS group, n=8), swimming endurance exercise a certain time; leptin obese group (OL group, n=8), injection of leptin lug/500g; sport and leptin obese group (SL group, n=8), let them have the leptin injections and swimming endurance exercise. Exercise group (OS) and sports leptin group (SL) began swimming at two in the afternoon every day, but the sport and leptin group (SL group) began inject the leptin after the exercise; the rats have the rest for one day at the seventh day every week, they movementde for30minutes the first day, and they had swimmed for45minutes the next day, they have the adaptive sports for60minutes at the third day,and then let them continue the60minutes for the last three days. They had sported increased by15minutes in the next three weeks each week,and the last week of105minutes a day. The leptin intravenous measurements,0.05ml the Leptin solution/100g weight,injected the leptin (OL) and sports leptin (SL) tendons every two in the afternoon. The last day,we measured the height, the liver, gastrocnemius. We measured lipase (LPS), hepatic lipase (HL), lipoprotein lipase (LPL), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activity; malondialdehyde (MDA) content by ELISA in rat liver and skeletal muscle.Results:(1)In the whole experiment, the weight of each group of obese rats were positive growth, the average growth rate of the O rats were faster than the OS group,OL group and SL group (P<0.05);(2) The liver-to-body ratio coefficient of the OS group, the OL group, the SL group should be higher than the O group, the liver-to-body ratio coefficient of SL group were lower than OS group and OL group, the liver-to-body ratio coefficient of the OL group was higher than the OS group, but these differences were not significant difference (P>0.05);(3) The LPL activity of the SL rats’ liver was higher than the OS group, and there was a significant difference (P<0.05);the LPL activity of the OL group was higher than the OS group, and there was significant difference (P<0.05),the LPL activtiy of SL group was higher than OL group(P<0.05); the HL activity of the intervention group rats’ liver have improved, the HL activity of the OL group is higher than the O group, and there was significant difference (P<0.05).(4) Compared with the activity of LPS of O group, OS group, OL group the group SL LPS activity have been reduced, and the OS group was very significantly reduced (P<0.01), SL group was very significant reduced (P<0.01), OL group was significantly reduced (P<0.05); Compared with the LPL activity of the group O, the OS group has been improved and there was a significant difference (P<0.05), while the OL group, the SL group has declined and there was no significant difference (P<0.05); Compared with the OS group, the LPL activity of the OL group and the SL group was low,and there was a significant difference (P<0.05); compared with the HL activity of the O group, the OS group was significantly reduced (P<0.05); copared with the HL activity of OS group, the OL groups and the SL group were high, and there was a significant difference (P<0.05);(5)We could see from the SOD activity data, compared with the O group, the intervention group (OS, OL, SL group) has reduced,the OL group and SL group was significantly decreased (P<0.05); we could see from the GSH-Px activity data of rats’ liver, compared with the OL group, SL group was decreased, and there was a significant difference (P<0.05); we could see from the MDA content data of rats’ liver, compared with the O group, OS, OL, SL group has increased, and there was a significant difference (P<0.05),and the OS group and OL group has increased(P<0.05);(6)We could see from the SOD activity data of rats’ muscle, compared with the O group, the OS group and the SL group have increased, and there was a significant difference (P<0.05); compared with the SL group,the OS group and OL group have been higher, and there was a significant difference (P<0.05); we could see from the GSH-Px activity data, compared with the O group,the OS group, OL group and the SL group have improved significantly, and the OL group has significantly improved (P<0.05), the SL group also has significantly improved (P<0.05);(7) We could see from the correlation data analysis,the SOD activity and MDA content significantly was related (|γ|>0.7, P<0.05) from the data of the O group; the LPL and SOD activity had a very significant correlation (|γ|>0.7, P<0.01)from the OS group data; from the the OL group data, LPS and LPL activity there was a significant correlation (|γ|>0.7, P<0.05), the activity of LPL and HL have significantly related (|γ|>0.7, P<0.05), GSH-Px activity and MDA content have significantly correlated (|γ|>0.7, P<0.05).Conclusions:(1) The effect is what of the swimming endurance exercise for obese rats through four weeks. First, the movement of the the obese rats weight was positive growth, but the growth rate was lower than the quiet group; Second, the effect of exercise on skeletal muscle lipid metabolism enzymes was inconsistent, the mechanism needs further study;Third, movement of liver cells caused some damage; Fourth, the exercise could improve the antioxidant capacity of skeletal muscle;Fifth, the LPL activity and the SOD activity of rat liver were negatively correlated.(2)The effect is what of leptin for obese rats. First, injection of leptin in obese rats’weight was positive growth; Second, leptin could enhance the lipid metabolism of the rat liver, but the liver lipid metabolism of movement was more than leptin;Third, leptin in obese rats could improve the lipid metabolism of bone muscle, but exercise could enhance the ability of lipid metabolism of skeletal muscle more; Forth, the leptin reduced the antioxidant capacity of the liver,and it caused some damage to the liver;the leptin could improve the antioxidant capacity of rat skeletal muscle; Fifth, it’s negative correlation between LPS and LPL activity in obese rat liver with leptin, LPL and HL activity was negatively correlated,GSH-Px activity and MDA content were negatively correlated; Sixth,it’s positively correlated between LPS and GSH-Px activity with leptin acting on obese rats skeletal muscle,the LPL activity and MDA content was negatively correlated,the HL and GSH-Px activity was positively correlated.(3) The effect is what of leptin for endurance exercise in obese rats. First, Leptin can improve the ability of lipid metabolism of rat liver; Second, the impact of leptin on rat liver lipid metabolism activity is inconsistent, the ability of the liver lipid metabolism mechanism needed further study, the effect of that the leptin acted on the endurance exercise were no consistent effect on lipid metabolism in rat skeletal muscle,it reduced fat would be not achieve a consistent role; Second, the leptin could reduce the antioxidant capacity of exercise obese rat liver, and the leptin increased the lipid peroxide of skeletal muscle, therefore, the leptin was not conducive to the antioxidant capacity; Fourth, endurance exercise and injection of leptin simultaneously could improve the anti-oxidative capacity of skeletal muscle.
