This dissertation consists of four chapters:The first chapter confirms the effect of different extracts of decoction of Qinzhu siwutang on dysmenorrhea model mouse. The second chapter describes the decoction of the chemical constituents of ethyl acetate extract parts of Qinzhu siwutang. The third chapter determines main chemical constituents and Single herbs from Qinzhu siwutang by RP-HPLC. The fourth chapter determines the change of baicalin, baiealein and wogonin contents in different decoctions prepared from Qinzhu siwutang by RP-HPLCTo observe the effect of different solvent-extracts of Qinzhu siwutang on dysmenorrheal model mouse induced by oxytocin. Acetidin and Ethylacetate parts of Qinzhu siwutang can obviously decrease the rate of the mouse twisting reaction induced by oxytocin, compared with the model group, they have significant or very significant difference(P<0.05, P<0.01). Ethylacetate part of Qinzhu siwutang are the most effective parts on dysmenorrhea.The compounds were isolated by column chromatography with silica gel and Gel Permeation Chromatography. Their structures were determined on the basis of physicochemical properties and spectroscopic analysis.17compounds were isolated and16were identified from Qinzhu siwutang. The identified compounds are tripalmitin,2(S)-5,7-dihydroxy-6-methoxyflavanone, β-sitosterol, wogonin, chrysin, benzoic acid, atractyle-nolide-Ⅲ,5,6’-dihydroxy-6,7,8,2’-tetramethoxyflavvone, trans-ferulic acid, formononetin, baicalein,5,7,3’,6’-tetramethoxy-8,2’-dimethoxy-flavone,2’,3,5,6’,7-pentahydroxy-flavanone,5,7,2’,6’-tetrahydroxyflavones, gallic acid, Wogonin-5-O-B-D-glucopside. All the compounds are isolated from Qinzhu siwutang for the first time. Wogonin and baiealein are the bioactive components of Qinzhu siwutang for treating primary dysmenorrhea.To determines main chemical constituents and Single herbs from Qinzhu siwutang by RP-HPLC. The results of RP-HPLC qualitative analysis showed that most of the main components is Baicalin, Baiealein and Wogonin. These results meant that the main components is flavonoids in Radix Scutellariae.At last, To determine baicalin, baicalein and wogonin contents in decoctions prepared from Qinzhu siwutang by RP-HPLC. The lowest content of baicalin is lack of White Paeony Root in Qinzhu siwutang, The content is40.01±0.03mg/g; The highest content is compatibility of Radix Scutellariae, Chen medicine and shi medicine, the content is56.48±0.10mg/g; The content of Qinzhu siwutang is42.46±0.04mg/g. The lowest content of baicalein is lack of Rhizoma Atractylodis Macrocephalae in Qinzhu siwutang, The content is8.87±0.07mg/g; The highest content is compatibility of jun medicine and Rhizoma Ligustici Chuanxiong, the content is16.22±0.04mg/g; The content of Qinzhu siwutang is9.97±0.09mg/g. The lowest content of Wogonin is lack of White Paeony Root in Qinzhu siwutang, The content is2.48±0.06mg/g; The highest content is compatibility of jun medicine and zuo medicine, the content is4.48±0.09mg/g; The content of Qinzhu siwutang is3.08±0.03mg/g. The method for determination of3flavonoids compounds in Qinzhu siwutang, the method can be used for rapid and convenient evaluation of the quality of Qinzhu siwutang. The highest content is Radix Scutellariae, the content of decoction of Qinzhu siwutang is among all decoctions. The results of RP-HPLC qualitative analysis showed that most of the3flavonoids compounds did change obviously in different decoctions. These results meant that some remarkable changes occurred in this compatibility.
