Objective:To observe the effect of total flavonoids of Dodder on dysmenorrhea with estradiol benzoate and the estradiol benzoate combined with ice water bath in rats,and to explore the therapeutic effect and characteristics of total flavonoids of Dodder on dysmenorrhea.Methods:The mice model was used,where estradiol benzoate caused the dysmenorrhea,except for the blank group,the mice in remaining groups were injected with estradiol benzoate for 7 successive days.On the 5th day after molding,the rats were administered for 3 days,after the last gavaged 40 min and the number of writhing in rats observed within 15 minutes after injection with oxytocin.Hemodynamic indexes,prothrombin time,thrombin time,and activated partial thrombin time were detected,the levels of PGF2a,PGE2 and NOS in uterine homogenate were measured and the changes in histomorphology of the uterus,thymus,liver and spleen were observed under light microscope.The rat model was used,where estradiol benzoate caused thrombin time were detected,the levels of hs-CRP in their serum,the levels of 6-keto-PGF1aa in plasma and the levels of PGF2a,PGE2 and NOS in uterine homogenate were measured;The expression of VEGFR-2 protein in the uterus was detected by Western-blot and the changes in histomorphology of the uterus,thymus,ovary and spleen were observed under light microscope.The mouse model was used,where estradiol benzoate combined with ice water bath caused the dysmenorrhea,except for the blank group,the mouse in remaining groups were immersed in ice water at 0?-1?every day,and injected with estradiol benzoate for 12 successive days.On the 8th day after molding,the mouse administered for 5 days,after the last gavage for 40 min and the number of writhing in rats was observed within 15 minutes after injection with oxytocin.Hemodynamic indexes,prothrombin time,thrombin time,and activated partial thrombin time were detected,the levels of?-EP?IL-6in their serum and the levels of AVP?ET-1?GSH-PX in uterine homogenate were measured,and the changes in histomorphology of uterus,thymus,ovary and spleen were observed under light microscope.The rat model was used,where estradiol injection with oxytocin.On the 13th day,except for the blank group,the rats in the model group,aspirin group,motherwort group,large,medium and small dosage dodder total flavonoid groups were injected intraperitoneally with hydrochloride adrenaline and stimulated with ice water,injected intraperitoneally with 10%chloral hydrate for anesthesia,and blood was collected from abdominal aorta.Hemodynamic indexes,prothrombin time,thrombin time,and activated partial thrombin time were detected,the levels of 5-HT,E2,LH,PROG,T and FSH in their serum and the levels of NO,PGE2 and PGF2a in uterine homogenate were measured,and the changes in histomorphology of the uterus,thymus,ovary,spleen,liver and kidney were observed under light microscope.Results:Through the study of induced dysmenorrhea in a mouse model of estradiol benzoate:Compared with the blank group,model group mice writhing latency significantly increased,decreased significantly,The level of PGE2 and NO in the uterine homogenate was significantly decreased,and the level of PGF2?was significantly decreased significantly increased,Mouse uterus,liver organ index was significantlyincreasedanditsspleen,thymusorganindexdecreased significantly.Compared with the model group,large,medium and small dose of Dodder total flavone group and aspirin group,motherwort group can improve the writhing times,improve latency,increased PGE2 and NO levels in uterus homogenate and reduce the levels of PGF2a,improved the pathological changes of related organizations.Through the study of induced dysmenorrhea rat model of estradiol benzoate:Compared with the blank group,the model group rats writhing latency significantly increased,decreased significantly,its serum hs-CRP significantly increased and the content of 6-keto-PGF1a in plasma decreased significantly,PGE2 and NOS level in uterus homogenate decreased significantly,PGF2aa and VEGF level increased significantly,significantly increased uterine and ovarian organ index,spleen and thymus index decreased significantly,significantly increased the expression of VEGFR-2 protein in uterine tissue of model group rats.Compared with the model group,large,medium and small dose of Dodder total flavone group and aspirin group,motherwort group can decrease the number of writhing rats,increase rats writhing latency,decreased PGF2aa and hs-CRP in uterine homogenate of alpha VEGF levels in serum,increase the content of 6-keto-PGF1aa in plasma,improve the pathological changes of tissues,decrease the expression of VEGFR-2 protein in the rat uterus.Through the study of estradiol benzoate combined with ice water bath in Dysmenorrhea mice:Compared with the blank group,the number of writhing in the model group was significantly increased,the latency was significantly decreased,and the levels of?-EP and IL-6 in the serum were significantly higher than those in the control group.The levels of AVP and ET-1 in uterine homogenate were significantly decreased,the level of GSH-PX was significantly decreased,the index of uterus and ovarian organ was significantly increased,the index of spleen and thymus was significantly decreased.Compared with the model group,all the Dodder total flavonoids groups,aspirin group and aspirin group and motherwort group decreased the number of writhing in mice,increased the latency of writhing in mice,increased serum?-EP,IL-6 and uterine Pulp AVP,ET-1 level,reduce the level of GSH-PX in uterine homogenate,improve the relevant histopathological changes.Through the study of estradiol benzoate combined with ice water bath in Dysmenorrhea rats:Compared with the blank group,the number of writhing in the model group was significantly increased,the latency was significantly decreased,the plasma viscosity,hematocrit,erythrocyte aggregation,rigidity and erythrocyte aggregation were significantly higher than those in the control group.The levels of5-HT,E2,T,P,LH and FSH in serum were significantly increased,and the levels of prothrombin time,partial thromboplastin time and thrombin time were significantly shortened PGE2,NO levels were significantly decreased,PGF2?levels were significantly increased,uterus,ovarian organ index increased significantly,spleen,thymus organ index decreased significantly.Compared with the model group,the total flavonoids of large,medium and low dose dodder can reduce the number of writhing,increase the latency of writhing,decreased the plasma viscosity,hematocrit,erythrocyte aggregation,rigidity and deformation index,increase the prothrombin time.The levels of PGE2 and NO in the uterine homogenate were increased and the levels of PGE2 and NO in the uterine homogenate were increased and the histopathological changes were improved.Conclusion:It is suggested that the Dodder total flavonoids of Cuscuta can improve the estradiol estradiol,the mice and the estradiol benzoate combined with the ice water bath,and the internal environment of the mice and improve the dysmenorrhea caused by the change of the hormone level and relieve symptoms of dysmenorrhea.
