Objective:In ramipril,for positive control medicine,observe the Yishenkanggranula on diabetic rats model of microalbuminuria and renal pathologicalmorphology,and evaluation of different concentration of kidney kang granuleson the curative effect of early diabetic nephropathy.Material and Method: SPF SD male rats,120randomly selected,109rats byintraperitoneal injection of chain urea with cephalosporins (STZStreptozotocin,53mg/kg) induced diabetic rats model.After72hours of orbitalvenous blood and measure blood sugar levels, blood glucose values or greatertendency for16.7L of rats were randomly divided into model group, positivedrug group, Yishenkang granula low-dose group, particles dose group, Yishenkanggranula high-dose group. For6weeks,8weeks urine were taken over the weekend,the content of Microalbuminuria was detected by Immunoturbidimetric Assay method.Dosing8weekend, anesthesia in rats after harvesting the left kidney cut, to10%formaldehyde fixed, make lens samples.Result and analysis:8weekend, then observe each group glomerularmorphology, size, mesangial matrix changes as a result of: Normal group ofglomerular structure, shape, size is normal, blood vessel, and the proportionof the bowman’s capsule cavity is normal, the proximal tubules and distal tubulewall thick, lumen size is normal; Model group rats glomerulus atrophy of glomus,renal lesion in cavity, ratio, the proximal tubules and distal tubule wall thickirregular, tube cavity sizes, swelling, there are a large number of inflammatorycells infiltration, compared with normal group kidney pathological structuraldamage is serious,it proves that diabetic nephropathy made film success; Chinesemedicine group and positive control medicine group compared weth model groupdamage slightly, Yishenkang granula and ramipril both can reduce the renal pathological damage early DN model role;6and8weekend over the weekend, themicroalbuminuria content of model group were significantly higher than in normalgroup (p <0.01), the6weeks I have diabetic nephropathy rats into film. The8weekend microalbuminuria content is significantly higher than model group6weekend, extended instructions with the course of the disease, the kidney damageis aggravating gradually; Positive drug control group and Chinese medicine groupmicroalbuminuria levels were significantly lower than the model control group(p<0.05), indicating ramipril and Yishenkang granula has reduced the role ofmicroalbuminuria; Chinese medicine groups in microalbuminuria protein comparedwith positive drug control group no significant difference (p>0.05), The effectof Yishenkang granula and ramipril is considerable; Comparison between theChinese medicine groups is no statistical significance, including betweendifferent Yishenkang granula dose groups do not exist the concentration-responserelationship; Chinese traditional medicine group of8weekend decreasedmicroalbuminuria is better than that of6weekend, shows that the longer thetreatment curative the better the effect.Conclusion:1.Early diabetes kidney disease animal model is successfully established.2.Yishenkang granula have the effect of prevention and treatment of earlydiabetic nephropathy by reducing microalbuminuria3.The curative effect Yishenkang granula of high, medium and low dose groupsis no difference.4.The prevention and treatment effect of Yishenkang granula and ramipril forearly diabetic nephropathy is considerable.5.Both Yishenkang granula and ramipril can reduce the early diabetic nephropathyrenal pathological damage.
