Research objective: To explore the relationship of MMP-3and COL9-A1inrabbit’s lumbar disc degeneration by establishing the rabbit model of lumbar discdegeneration, and testing early expression of MMP-3and COL9-A1Material and method: Checking42health New Zealand white rabbits,its weightis2Kg and age is4month. The rabbits were randomly divided into2groups (modelgroup, normal group). Model group was made model of lumbar disc degeneration,andControl group was not processed.4、8、12weeks after the operation, five rabbitsrandomly selected each cycle,which had X一ray and MRI examination, were killed andwere observed the disc’s change with naked eyes and histological change with HEstaining, to make sure the disc degeneration and its level.The expression of MMP-3ofmodel and control group’s discs was tested by immunohistochemisty SP and ELISA andthe expression of COL9-A1of model and control group’s discs in different time Points byimmunohistochemisty SP and RT-PCR. The results use SPSS17.0software package to Ttest, Spearman’s rank Correlation (significant level of a=0.05).Based on the results,therelationship between MMP-3and COL9-A1in lumbar disc degeneration.is furtherexplored.Result:1.4、8、12weeks after surgery, the level of lumbar disc degeneration of themodel group increased gradually. The control group had no apparent change1. The results showed there were more strong express of MMP-3in model groups thancontrol groups. On the contrary,there were less strong express of COL9-A1in modelgroups than control groups (p<0.05).2. There was certain negative relation protein expression of MMP-3and COL9-A1inmodel groups and control groups (r=-0.732). Conclusion:1. Rabbit lumbar intervertebral disc degeneration model was established byAnnulus fibrous puncture.2. The level of express of MMP-3in intervertebral disc degeneration increased.With thepassage of time,the level of express of MMP-3in intervertebral disc degenerationincreased gradually.3.The level of express of COL9-A1in intervertebral disc degeneration decreased。Withthe passage of time,the level of express of COL9-A1in intervertebral disc degenerationdecreased gradually.4.There was certain negative relationship between MMP-3and COL9-A1in lumbarintervertebral disc degeneration.5.The research of MMP-3and COL9-A1in lumbar disc degeneration supply a theoreticalbasis to biologic treatment and Diagnosis for lumbar intervertebral disc degeneration.
