Objective:(1)Establishing rats models of middle cerebral artery occlusion (MiddleCerebral Artery Occlusion, MCAO),to observe the expression of Tong Du Tiao Shenacupuncture pretreatment on cerebral ischemia reperfusion rats related miRNAs (microRNAs, miRNAs), Matrix Metallo Preteinases-9(Matrix Metallo Preteinases-9,MMP-9),aquaporin4(aquaporin4, AQP4).(2)To observe the the relevance of Tong DuTiao Shen acupuncture pretreatment on cerebral ischemia reperfusion rats relatedmiRNAs,MMP-9,AQP4expression and ischemic preconditioning.(3) To observe thedifferences of cerebral ischemia reperfusion rats related micrornas,MMP-9,AQP4expression between Tongdu Tiaoshen acupuncture and moxibustion in the sametreatment.(4) Comparing the edema protective effect between acupuncture andmoxibustion pretreatment on cerebral ischemia reperfusion rats,to provide laboratorybasis for clinical.Method: Selection of clean level healthy male Wistar rats130(3-4month old), weightof250g-300g, after one week adaptive breeding began to experiment. Rat breedingenvironment humidity of45-65%, the temperature is22to25℃.Using computerrandom number method divided rats into A:acupuncture pretreatment group (30),B:moxibustion pretreatment group (30), C:aspirin pretreatment group (30), D:modelgroup(30) and E:blank control group (10).Normal group beheaded for brain and save the brain tissue; Model group animalsrecord the neurobehavioral score after reperfusion24h,then beheaded for brain and savethe brain tissue; EA group, electro-acupuncture treatment once a day,20minutes at atime,7d for a course of treatment; moxibustion pretreatment group: use moxa-cigar tosuspended burning once a day,20minutes at a time,7d for a course of treatment;Aspirinpreconditioning group: Aspirin10mg/kg, dissolved in2ml normol saline to lavageonce a day,7d for a course of treatment; To produce cerebral ischemia reperfusion model afterthe treatment, record the neurobehavioral score after reperfusion24h, then beheaded for brain and save the brain tissue;Take the each group brain tissue for histologicalobservation, to observe the expression related miRNAs,MMP-9,AQP4in cortex.Results:(1)The neurobehavioral scores of model group,aspirin group, moxibustion andelectric acupuncture group were significantly improved. Compared with the normalgroup, each group neurobehavioral score difference had statistical significance (P<0.01);Compared with model group, the pretreatment groups scores difference had statisticalsignificance (P<0.01); Compared with aspirin group and moxibustion group, electricacupuncture group score difference was statistically significant (P<0.01). The resultsshowed that:1),MCAO rats model success;2), aspirin pretreatment, moxibustionpretreatment and electric acupuncture pretreatment can reduce ischemia reperfusion ratsneurobehavioral scores, and electric acupuncture preconditioning effect was better thanthat of aspirin and moxibustion pretreatment.(2) The model group, aspirin group, moxibustion and electric acupuncture group brainwater content were significantly improved. Compared with the normal group, eachgroup brain water content difference had statistical significance (P<0.01); Comparedwith model group, the pretreatment groups scores difference had statistical significance(P<0.01,P<0.05); Compared with aspirin group and moxibustion group, electricacupuncture group score difference was statistically significant (P<0.01,P<0.05). Theresults showed that: aspirin preconditioning, moxibustion pretreatment and electricacupuncture pretreatment can reduce ischemia-reperfusion rats brain water content, andelectric acupuncture preconditioning effect was better than that of aspirin andmoxibustion pretreatment.(3)Compared with the model group, the cerebral infarction volume of pretreatmentgroup were significantly lower. Compared with the model group, cerebral infarctionvolume of various pretreatment group difference had statistical significance(P<0.01,P<0.05); Compared with aspirin group and moxibustion group, electric acupuncture group score difference was statistically significant (P<0.01,P<0.05). Theresults showed that: aspirin preconditioning, moxibustion pretreatment and electricacupuncture pretreatment can reduce ischemia-reperfusion rats cerebral infarctionvolume,and electric acupuncture preconditioning effect was better than that of aspirinand moxibustion pretreatment.(4)Model group, aspirin group, moxibustion and electric acupuncture group miRNA290and miRNA494relative expression were significantly reduced. Compared with thenormal group, each group miRNA290, miRNA494relative expression differences werestatistically significant (P<0.01); Compared with model group, the pretreatment group,all the relative expression difference had statistical significance (P<0.01); Comparedwith aspirin group and moxibustion group, electric acupuncture pretreatment relativeexpression difference was statistically significant (P<0.05). The results showed thataspirin preconditioning, moxibustion pretreatment, electric acupuncture preconditioningcan improve the relative expression of miRNA290and miRNA494of ischemiareperfusion rats, electric acupuncture pretreatment and moxibustion pretreatment effectwas more significant.(5)The model group, aspirin group, moxibustion and electric acupuncture groupmiRNA664relative expression increased significantly. Compared with the normalgroup, each group miRNA664relative expression differences were statisticallysignificant (P<0.01); Compared with model group, all the pretreatment groups relativeexpression difference had statistical significance (P<0.01); Compared with aspiringroup and moxibustion group, electric acupuncture group relative expression differencewas statistically significant (P<0.01,P<0.05). The results showed that aspirinpreconditioning, moxibustion pretreatment and electric acupuncture preconditioning canimprove the relative expression of miRNA664of ischemia reperfusion rats, electricacupuncture pretreatment and moxibustion pretreatment effect is more significant. (6)The model group, aspirin group, moxibustion and electric acupuncture group therelative expression of MMP9and AQP4were significantly improved. Compared withthe normal group, each group miRNA664relative expression differences werestatistically significant (P<0.01); Compared with model group, all the pretreatmentgroups relative expression difference had statistical significance (P<0.01,P<0.05);Compared with aspirin group and moxibustion group, electric acupuncture grouprelative expression difference was statistically significant (P<0.01). The results showedthat aspirin preconditioning, moxibustion pretreatment, electric acupuncturepreconditioning can reduce the relative expression of MMP9and AQP4ofischemia-reperfusion rats, the effect of electric acupuncture pretreatment was moresignificant.Conclusion:(1)Tongdu Tiaoshen acupuncture preconditioning can significantly reduceneurobehavioral scores and cerebral water content of cerebral ischemia reperfusion rats,reduce the cerebral infarction volume, is an effective method to prevent cerebralinfarction.(2)Tongdu Tiaoshen acupuncture preconditioning can obviously increase the relativeexpression of miRNA290and miRNA494of cerebral ischemia reperfusion rats, reducethe relative expression of miRNA664, induce ischemic tolerance, formation of cerebralprotection, this may be a general overseers of the function of Tongdu Tiaoshenacupuncture preconditioning cerebral ischemia-reperfusion mechanism.(3)Tongdu Tiaoshen acupuncture pretreatment can effectively reduce the relativeexpression ofAQP4and MMP9of ischemia-reperfusion rats, relieve cerebral edema.(4)Tongdu Tiaoshen acupuncture pretreatment through regulating the expression ofmiRNA290, miRNA494and miRNA664to lower relative expression of AQP4andMMP9, induce ischemic tolerance, relieve cerebral edema. This may be the cerebralprotection mechanism of Tongdu Tiaoshen acupuncture pretreatment。(5)Tongdu Tiaoshen acupuncture preconditioning can improve cerebral ischemiareperfusion, formation of cerebral protection, and Tongdu Tiaoshen acupuncture preconditioning effect is better than moxibustion pretreatment.
