Background Sepsis is a clinical severe case, and it is the main cause of multiple organ dysfunction syndrome (MODS). Acute lung injury is common in these cases. It’s mortality is still very high. Most of all think the nature of acute lung injury is excessive, out of control of inflammation now. A variety of cytokines play an important role. A lot of tumor necrosis factor-a and interleukin1, platelet activating factor were generated when Sepsis occurred. It is reported that stem and leaf Ginsenosides have protective effects on lung in septic rats.Objective This study aims to study Effects of Stem and Leaf Ginsenoside to TNF-α、IL-1and PAF level of acute lung injury rats to observe it has or no protection to acute lung injury and discuss the possible protection mechanism.Methods40male adult SD rats were randomly divided into four groups:control group, sepsis model group, Ginsenoside group, Methylprednisolone group.Control group. Sepsis model group were injected with NS (5ml. kg-1), Ginsenoside group were injected with90%Ginsenoside (100mg. kg-1. ml-1), Methylprednisolone group were injected with Methylprednisolone (40mg. kg-1. ml-1). Sepsis model were made by cecal ligation and puncture (CLP) after two hours;control group without CLP,only open enterocoelia.The aorta abdominalis blood was puncted for sample after12Hours of the operation.The arterial gas was examined.Rest of the blood samples were centrifugated in3000r/min for15mins,the upper layer serum was frozen at-70℃for testing the levels of TNF-α、IL-1and PAF. The upper lobe of left lung tissue of rats were fixed by4%formalin for12hours, routine dehydration, transparent, soaking in candle, paraffin imbedding, serial slice up, taking HE dyeing and airproofing. Observing under microscope and taking pictures. Making tissue homogenate of inferior lobe of left lung. The level of serum TNF-α IL-1and PAF were measured by double double antibody enzyme-linked immune sorbent assay (ELISA) expermentation.The content changes of inferior lobe of left lung tissue were measured by same way too.Results1. The arterialoxygen partial Pressure (PaO2) of sepsis model group. Ginsenoside group, Methylprednisolone group rats were lower than control group(P<0.05),PaO2of Ginsenoside group and Methylprednisolone group were higher than sepsis model group (P<0.05).2. Histopathological microscope observed:control group lung tissue structure were clear, no congestion and oozy; sepsis model group lung microvascular had hyperaemia, micro thrombosis, oozy obvious,a lot of inflammatory cells infiltrated; Ginsenoside group histopathologic changes were similar to the sepsis model group, But damage was much lighter; The damage of Methylprednisolone group was lighter than Ginsenoside group.3. The level of TNF-α、IL-1and PAF in sepsis model group、Ginsenoside group and Methylprednisolone group rats were higher than control group(P<0.05) in serum and lung tissue homogenate. Their level in Ginsenoside group and Methylprednisolone group were lower than sepsis model group (P<0.05).Conclusions1. Injection with Stem and Leaf Ginsenoside may increase PaO2of septic rats and lighten acute lung injury. 3.The possible mechanism of protective effect on sepsis-induced acute lung injury is inhibiting inflammation. Stem and Leaf Ginsenoside can reduce the level of TNF-α、IL-1and PAF to restrain inflammation.
