Ginseng is the root of panax ginseng,which is a rare Chinese herbal medicine. As one of "Northeastern Three Treasures", it is on par with velvet, mink, and is of great medical activity value. The most important medicinal active ingredient in ginseng is ginseng saponin, the second is ginseng polysaccharide.Due to the high price and limited yield of ginseng, besides?root of Panax ginseng and panax ginseng stem and leaf contain the same medicinal active components, this experiment took panax ginseng stem and leaf as material to extract ginseng saponins and ginseng polysaccharide. The experimental contents are as follows:1.Optimal extraction process of ginsenoside in the stem and leaf of panax ginseng had been ensured by several single factor experiments and orthogonal experiment, results showed, optimal extraction process was ethanol 30%,80?hot water, reflux extraction 2 times,4.5h.2.Polymeric adsorbent XDA-7 was taken as optimal ginseng saponin separation material, which was selected from several macroporous adsorptive resins by static and dynamic test, then selected the best decolorization resin D762 from a variety of resins of different properties,to obtain the best purification products of the ginseng saponins.3.Resin multi column method combined with silica gel column chromatography was applied to purification separation of ginsenoside monomer Rgl and Re, ratio of elution agent in silica gel column chromatography was ensured by experiment: chloroform:methanol:ethylacetate=8.5:1:0.5,could accurately separate ginsenoside monomer Rgl and Re in accurate time.4.Appropriate thin layer chromatography was ensured to qualitatively analyze ginsenoside monomer Rgl and Re, and HPLC chromatography quantitatively to analyze ginsenoside monomer Rgl and Re.5.Optimal extraction process of Polysaccharide from ginseng stem and leaf had been ensured by several single factor experiments and orthogonal experiment,then refined the extracted polysaccharide to obtain ginseng polysaccharide purification products.
