Object:To observe the effect of sufentanil on contration and superoxide of thoracic arota isolated from rats with diabetes mellitus and investigate its mechanism,provide certain reference for clinical application of sufentanil on diabetic patients in the operation.Methods:There were8diabetes mellitus rats and8normal rats, every male rats were decapitated and their thoracic aorta were isolated. Aortic ring obtained from normal rat was randomly divided into4groups(n=8each):physiological saline control group(group NC), sufentanil group1(group NS1), sufentanil group2(group NS2), sufentanil group3(group NS3). Rings from DMR were randomly divided into4different groups (n=8):physiological saline control group(group DC), sufentanil group1(group DS1), sufentanil group2(group DS2), sufentanil group3(group DS3).Vitro vascular experimental method were used to observe the effect of different concentrations of sufentanil on the norepinephrine-induced contraction of thoracic aorta from diabetic rats and the content of NO, MDA and SOD on the vascular ring.Results:1) Group DC compared with the Group NC, the NE reduced contraction of the vascular ring was significantly. increased,NO content of vascular ring was lower,MDA content was higher and the SOD activity was lower(P<0.05).2) Compared with the Group NC, the vascular ring contraction in the group NS1was no obvious change,the content of NO,MDA and SOD on vascular ring also had no significant difference (P>0.05);which in the Group NS2and NS3, contraction were reduced, NO content of vascular ring was higher (P<0.05), but the content of SOD and MDA had no significant difference (P>0.05).3) Compared with the Group DC, contraction of the vascular ring in group DS1, DS2and DS3were reduced,NO content and SOD activity in the vascular ring was increased and MDA content was reduced (P<0.05).4) Compared with the Group NS3, the vascular systolic amplitude in the Group DS3were decreased significantly,NO content of vascular ring was lower,MDA content was higher and the SOD activity was lower(P<0.05).Conclusions:These results indicate that the contractile function of thoracic aorta in DMR was increased. Sufentanil can inhibit NE-induced contraction and change the NO,MDA and SOD of thoracic aorta isolated from rats with Diabetes mellitus in a concentration-dependent manner.
