ObjectiveToinvestigate the effect of sufentanil on norepinephrine(NE)-induced contraction of thoracic aorta isolated from type2diabetes mellitus rats (T2DR)MethodsEight male T2DRand eight male normal rats(NR) were used in the study. The rats were decapitated and their aorta rings were suspended for tension measurement device.The aortic rings obtained from T2DMRand NR were divided into4groups(n=8each):control group and3sufentanil groups(7×10-11and2×10-10and1×10-9mol·L-1)The contraction of aortic rings in response to NE in the absence and3concentrations of sufentanil was recorded and NO, NT-llevels was measured.Results(1) Compared with the Group NC, the amplitude of NE-induced contraction of the vascular ring was significantly increased in Group DC (P<0.05);and its NO content was decreased, ET-1concentrate was increased (P<0.05)(2)Compared with the Group NC, The amplitude of NE-induced contraction of thoracic aorta rings was significantly greater, their NO levels was higher and ET-1levels was lower than in group NS1,NS2,NS3(P<0.05);and their NO contents were increased, ET-1concentrate were decreased (P<0.05)(3)Compared with the Group DC,the amplitude of NE-induced contraction of thoracic aorta rings was significantly greater in group DS1,DS2,DS3,(P<0.05);and their NO contents were increased, ET-1concentrate were decreased (P<0.05)(4) Compared with the Group NS3, the amplitude of NE-induced contraction of thoracic aorta rings was significantly greater in groupDS3(P<0.05); and its NO content was decreased, ET-1concentrate was increased (P<0.05)ConclusionSufentanil can inhibit NE-induced contraction of thoracic aorta isolated by increasing NO, reducing ET-1in a concentration-dependent manner.
