Objective:To observe the effects of Jianpi Xiaozhi Formula on the expression changes ofweight, FBG, FINS, HOMA-IR, blood lipid, APN, TNF-α, CRP, serum chemerin andchemerin of perirenal fat and pancreas chemerin in model rats with type2diabeticdyslipidemia, and explore its mechanism of action.Methods:Sixty male SD rats were divided into2groups randomly, ten in blank controlgroup and fifty in diabetes group. Rats in the diabetes group were given intraperitonealinjection of STZ (27mg/kg) once only after four-week feed of fodder rich in highglucose and fat, so that some of the pancreas cells were damaged and hyperglycemiawas caused. All rats were fasted from food but allowed to intake water for twelve hourswith20%glucose solution force-feeding by the scale of2g/kg for body weight twoweeks later. OGTT experiment was operated and the diagnostic criteria were:①fatrats were20%heavier than those in blank control group.②TC, TG, HDL-C, LDL-C inblood serum of rats were determined and their contents were not in different levelsbefore the modeling. Whereas TC, TG, LDL-C were much higher than those in blankcontrol group (P<0.05), while HDL-C lower significantly (P<0.05).③OGTTexperiment was executed and the blood glucose higher than7.0mmol/L and/or11.1mmol/L (blood extracted in caudal vein and tested with glucometer) in0minand/or120min respectively could be counted as a success in modeling. Those rats withfailure to be modeled were again injected with STZ solution by the scale of27mg/kg,and then forbidden from food but allowed water for twelve hours again one week later.OGTT experiment was carried out and that the standards were met meant a success ofmodeling. Those rats failed to be modeled were excluded and the left were divided intofour groups randomly: model control group, Jianpi Xiaozhi group, simvastatin group and Jianpi Tating group with ten rats in each group. Rats in prescription group of JianpiXiaozhi group, simvastatin group and Jianpi Tating group were fed with relative drugs,while blank control group and model control group were given saline by the scale of5ml/kg/d. Each group was fed in continuous four weeks and the behavior, state of spiritand weight of each group were observed. Kill the rat by collection the blood fromabdominal aorta to acquire the blood, perirenal fat and pancreatic tissue into differenttube after the lasting gavages for four weeks. And then separate the serum and thetissue for testing the expression levels of FPG, FINS, TC, TG, HDL-C, LDL-C,TNF-α,CRP, APN, ALT, CR, serum chemerin, chemerin of perirenal fat andpancreatic tissue.Result:1. The weight of rat in model control group is higher than those in the blankcontrol group after modeling; the difference is significant (P<0.01). The weight of ratin Jianpi Xiaozhi group, simvastatin group and Jianpi Tating group are lower thanthose in the model control group after treating, the difference is significant (P<0.01orP<0.05).2. Compared with the rats in blank control group, the levels of FPG, FINS,HOMA-IR, TC, TG, LDL-C, TNF-α,CRP, serum chemerin, chemerin of perirenal fatand pancreatic tissue are increased significantly in the groups with diet and injectioninduced, but the level of HDL-C and APN are obviously decreased (P<0.01or P<0.05).3. Compared with the model group, the levels of FPG, FINS, HOMA-IR, TG,LDL-C, TNF-α,CRP, serum chemerin, chemerin of perirenal fat and pancreatic tissueare decreased significantly in the Jianpi Tating group, the simvastatin group and theJianpi Xiaozhi group, but the level of APN is obviously increased (P<0.01).thechange levels of TC and HDL-C in Jianpi Xiaozhi group and HDL-C in simvastatingroup are not obviously, it has no statistical significance (P>0.05). 4. Compared with the simvastatin group, the levels of FPG, FINS, TG, LDL-C,TNF-α,CRP, serum chemerin, perirenal fat tissue chemerin are decreased significantlyin the Jianpi Tating group, and the level of APN is obviously increased (P<0.01or P<0.05), but the change levels of TC, HDL-C and pancreatic tissue chemerin are notobviously, it has no statistical significance (P>0.05).Conclusion:Diet combined with low-dose STZ injection can successfully established type2diabetic dyslipidemia rat model. The levels of FPG, FINS, TC, TG, HDL-C, LDL-C,TNF-α,CRP, APN, serum chemerin, chemerin of perirenal fat and pancreatic tissuehave been improved significantly after the therapy of Jianpi Xiaozhi Formulacombined with simvastatin in type2diabetic dyslipidemia rat, and the curative effectof combined therapy is better than Jianpi Xiaozhi Formula or simvastatinsole treatment. The mechanism of those changes may be related to the lower levels oflipid peroxides, the reduction of inflammatory cytokine production, the decreaseexpression of serum chemerin and tissue chemerin, the improvement of insulinresistance and the regulation of immune function.
