Objective1To discuss the different methylation rates between Runx3gene and Gst-π gene in gastric carcinoma and adjacent tissues, and to investigate the significance of RUNX3gene and Gst-π gene in the carcinogenesis and progression of gastric carcinoma.2To discuss the relationship between the methylation of Runx3genes and Gst-π gene promoter with the clinical features of gastric carcinoma.3To discuss the relationship between the methylation of Runx3genes and Gst-π gene promoter with syndrome differentiation of "gastric carcinoma" in TCM.Methods1Collected30cases of primary gastric carcinoma tissues and their adjacent tissues(normal tissues away from carcinoma tissues edge>5cm) in general surgery of General Hospital of Guangzhou military command of PLA from March2011to November2011. all cases were confirmed by endoscopic biopsy and didn’t have chemotherapy or radiation therapy before surgery. then postoperative tissue samples confirmed by paraffin pathology diagnosis.2All patients diagnosed "gastric carcinoma" by TCM syndrome differentiation. Then used Chinese medicine’s "inspection, listening and smelling examination, inquiry, palpation" to syndrome differentiation.3Using methylation-specific PCR (MSP) to detect the methylation of RUNX3gene and Gst-π promoter in the specimens from gastric carcinoma patients (tumor tissues, adjacent tissues).4Used statistics software SPSS13.0to analyse above indexes’ statistical significance. Results1The methylation of Runx3gene in tumor tissues was70.0%(21/30), and16.7%(5/30) in the adjacent tissues, they had significant variation (P<0.05).2The methylation positive rate of Runx3gene has positive correlation with gastric carcinoma’s clinical stages and lymph node metastasis (P<0.05), but has no correlation with age, sex, differentiation degree (middle or low), infiltrating degree(P>0.05).3The methylation of Gst-π gene in tumor tissues was56.7%(17/30), and26.7%(8/30) in the adjacent tissues, they had significant variation (P<0.05).4The methylation positive rate of Gst-π gene has no correlation with gastric carcinoma’s clinical stages and lymph node metastasis, age, sex, differentiation degree (middle or low), infiltrating degree (P>0.05).5There is no relationship between Runx3gene and Gst-π gene in gastric carcinoma.6Syndrome of (blood) stasis in the stomach collateral (36.7%,11/30)and syndrome of incoordination beteeen liver and stomach (33.3%,10/30)were a majority of the syndrome differentiation of gastric carcinoma, they had no statistically significant between each other(P>0.05), but had statistically different with other syndrome types (P<0.05).7There were no statistically significant differences of the methylation rate of Runx3gene in syndrome differentiations of gastric carcinoma of TCM.8There were no statistically significant differences of the methylation rate of Gst-π gene in syndrome differentiations of gastric carcinoma of TCM.ConelusionHigh frequent hypermethylation of Runx3gene and Gst-π gene is mostly cancer specific(P<0.05), and the methylation of Runx3gene and Gst-π gene promoter correlated with the carcinogenesis and progression of gastric carcinoma. Study their’s methylation may have positive evaluations in early screening, prevention, cure and prognosis of gastric carcinoma. In guangdong, the surgery patients with gastric carcinoma are a majority of syndrome of (blood) stasis in the stomach collateral and syndrome of incoordination beteeen liver and stomach in syndrome differentiation of TCM.
