4-vinyl-2,6-dimethoxyphenol (Canolol) Suppresses Gastric Tumor Lesions In K19-C2mE Transgenic Mice

Objective: The purpose of our study was to evaluate the effectiveness ofcanolol for inhibition of chronic gastritis and gastric carcinogenesis inK19-C2mE transgenic mice.Methods: Eighty K19-C2mE mices,6weeks old, were used in this study.The eighty mices were divied into two groups (Control and Canolol-treated):[Group Control, n=40]: K19-C2mE, untreated animals;[Group Canolol-treated, n=40]: K19-C2mE, concentration of0.1%canolol in drinking watertreated animals. Animals were killed at52weeks of age for midtermexamination. And then weight curve, incidence of gastric tumor, gastric grossmorphology, pathologycal HE staining, immunohistochemistry, as well as theexpression of tumor necrosis factor-a (TNF-α) and cyclooxygenase-2(COX-2)mRNA were be analyzed. At last, we used the statistical analysis software toanalyse the impact of canolol on K19-C2mE transgenic miceResults: The incidence of gastric tumor was markedly reduced in thecanolol-treated group [41.2%(14/34)] compared to the control group[77.8%(28/36)](p <0.05). And the inflammatory responses of glandular mucosa wasalso reduced. Meanwhile compared to the control group, the tumor size wassignificantly reduced in the canolol-treated group. Immunoreactivity againstCOX-2and EP2was evident in the control groups, and H-scores in the canolol-treated groups were significantly lower than in the control groups.Furthermore real-time quantitative of PCR reflected the expression of tumornecrosis factor-a (TNF-α)、microsomal PGE synthase (mPGES)-1, mPGES-1and cyclooxygenase-2(COX-2) mRNA in the gastric mucosa weresignificantly lower in canolol-treated groups.Conclusion: Canolol can significantly reduce the incidence of gastrictumor, and also can effectively inhibit mucosal inflammation. Furthermorecanolol can suppress induction of mRNAs for inflammatory cytokines ofCOX-2, mPGES-1and TNF-α.
