Objective The experimental study aims to confirm the effectiveness of rhubarb by umbical application on slow transit constipation in mice, thus to provide experimental evidence for the application of the navel therapy clinically.Methods60healthy ICR mice, were randomly divided into five groups equally, which were the normal control group, the model control group, the gavage group, the navel therapy group and the positive drug control group, male and female, in separate cage. In the navel therapy group, rhubarb cake was compressed on ShenQue acu-point (1g/d),the gavage group were given rhubarb liquid, the positive drug control group were treated with mosapride citrate solution by gavage administration, the normal control group and model control group were given distilled water, delivery volume is0.1mL/10g, once a day. Each mice was in accordance with the above method of treatment for6days consistently, at day7, all mice except the normal control group were given compound diphenoxylate suspension by gavage administration to establish STC model. To compare the defecating function of the two routes of administration on STC mice, each group of mice were given instructive ink thirty minutes after modeling. Then began to record the first defecation time, amount of black stool in6hours and fecal water content index. To compare the intestinal motility of the routes of administration on STC mice, the above mice were feeding without drug for one month, then the mice were re-decided and re-administration according to the metood described above. In the seven day each each group of mice were given instructive ink thirty minutes after modeling. Thirty minutes later, picking each mouse’s eyeball for blood, collecting blood sample and then dissecting the mice. After that, measuring small intestinal propulsion rate, detecting the NO content in serum and intestinal tissue with nitrate reductase method.Results In the experiment of observing the defecation function, experimental results show that the time of first black stool of the model control group is242.33±50.78min, the amount of black stool in6hours is7.83±3.79granule, moisture rate was5.96±2.15%.while the normal control group was170.33±57.54min,13.92±5.57granule and6.59±0.87%5which indicates that,compared with the normal group, the time of first black stool of the model control group was prolonged apparently and the amount of black stool in6hours decreased significantly, the difference was statistically significant (P<0.05), which proves that the STC mode was established successfully. The time of first black feces of the navel therapy group was197.50±51.82min, the total black grain in6hours was14.58±6.20granule, moisture rate was7.64±0.80%, while the gavage group were185±39.90min,11.75±5.74granule,9.78±2.17%.The experimental results show that, both of the gavage group and the navel therapy group’s first time of black stools were shortened, their total black grain in6hours increased, moisture rate was significantly higher, compared with the model group, the difference was statistically significant (P<0.05), indicating that rhubarb by the two routes of administration improve defecation function on STC obviously. For the comparison of the defecation function between two groups, the navel therapy group’s time of first black stool was shorter than the gavage group, its total black grain in6hours was more than the gavage group, but its moisture rate was lower than the gavage group. In the experiment of observing the effcct on the intestinal motility, the intestinal propulsive rate of the navel therapy group was60.94±17.92%, the level of NO in serum was121.57±46.84umol/L, the level of NO in tissue was98.86±61.52pg/g.While the gavage group was51.42±17.32%,124.79±56.56umol/L,150.23±46.89pg/g, the model control group was45.14±9.87%.168.83±47.55umol/L,150.23±46.89pg/g.The experimental results show that compared with the model group,the navel therapy group’s small intestinal propulsion increased (P<0.01), while the gavage group has no significant difference (P>0.05). The level of NO in serum and tissue both in the navel therapy group and the gavage group decreased significantly, compared with the model group (P<0.05).Conclusions The navel therapy of rhubarb can effectively improve the slow transit constipation defecation function. However, there is no significant difference between the the navel therapy and the gavage administration of the effect on slow transit constipation mice. In a word, the experimental results show that, the effectiveness of rhubarb by two different routes of administration on constipation has no significant difference. Due to that the navel therapy has the advantages of simple operation, definite curative effect, low cost, safe and easy to popularize, while oral administration of rhubarb for long time will result in a series of side effects, such as intestinal motility decreased, cathartic colon, melanosis coli and so. Thus, this experimental provide the basis for the prevention and treatment of STC by applying the navel therapy, and indicate the navel therapy of rhubarb has good generalization.
