Plantain the traditional Chinese herbal medicines is the seeds ofPlantaginaceae plant Plantain. According to Chinese Pharmacopoeia[1]thePlantain Seed need to be fried to have the focal spot before the raw PlantainSeed used. After testing we found the Plantain Seed riches of polysaccharides,proteins[2-3], choline and many other ingredients. There are studies found thatplantain seed polysaccharide (PSP) has the anti-aging, stop diarrhea, anti-hypercholesterolemic, lowering blood lipid pharmacological effects, but fewof anti-atherosclerosis. The study was aimed to investigate the effect and themechanism of the PSP how to resist the atherosclerosis. As the traditionalChinese medicine Plantain Seed has the characteristics of low cost and lowtoxicity, we will provide a more scientific and comprehensive theory of PSPfor clinical applications.Objective: Aimed to explore the anti-atherosclerosis effects and themechanism of PSP in high fat diet mice[4], we test the content of enzyme,serum, liver lipid levels. Then we find PSP has the effects of lowering bloodlipid, anti-oxidation and enhancing body immunity.Method:1The50Wistar rats weighing from170to200g, randomly divided intofive groups, each group get10rats. NC: normal control group; HL: high-fatdiet group.HLL: high-fat diet+PSP low dose; HLM: high-fat diet+PSPmiddle dose.HLH: high-fat diet+PSP high dose. The rats of the normalcontrol were fed with the normal diet; the other rats were fed with the high fatdiet. The rats of PSP group were gaveled the Plantain Seed Polysaccharides100mg/kg,200mg/kg,400mg/kg everyday, the rats of NC and the HL weregaveled2ml/kg water everyday.2All animals were free access to food and water, during the12weeks, they got weights every week.3We select blood by hit the inner cantus at the1st, the3rd, the8th, the12th week. Viewing the serum TG, TC, HDL-C, LDL-C and testing the AI.4After12weeks, collect arterial blood, aortic vascular and the liver tocalculate the organ coefficient. Cutting part of aorta and liver and the makethem into HE samples to observe the plaques and lesions.5Detecting the SOD and MDA in the serum and liver homogenate.6Testing the content of ALT, AST, LPL, and HL in serum by using thebiochemical kit.7Viewing the content of HMG-CoA in liver of rats by using the ELLISAmethods.Results:1There was a not significant difference between different groups whentesting the body weight and the serum lipid before the experiment. After12weeks the body weight of HLM and HLH group rats was significantly lowerthan the M group (P<0.05).2Compared with the NC group the content of TC, LDL-C, HDL-C fedwith the high-fat diet rats were significantly higher; The TC, LDL-C, HDL-Cof HLM and HLH group were significantly lower than the HL Group (P<0.05),but the HDL-C levels were not significantly elevated.3By observing the hepatic and aortic HE staining under an opticalmicroscope, we found the infiltrating cells in blood vessel wall and liver ofPSP groups were lower than the HL group.4Compared with the HL group the activity of SOD in serum and liver ofHLM and HLH groups were significantly improved (P<0.05); the content ofMDA in serum and liver were significantly decreased (P<0.05).5The serum ALT, AST of HLM, HLH group rats compared with the NCgroup were significantly decreased (P<0.05); the content of serum HL inHLM, HLH groups was significantly increased compared with the HL group(P<0.05).6The HLM, HLH groups’ content of liver HMG-CoA was lower compared with the HL group (P<0.05).Conclusion:1The Plantain Seed Polysaccharides can reduce the blood lipid levels,which play the inhibition effect in the early atherosclerotic lesion.2Plantain seed polysaccharides can reduce peroxide levels in serum andliver, and then play an important role in delaying the formation ofatherosclerosis.3The Plantain Seed Polysaccharides can play the anti-atherosclerosis byincrease the body’s immunity.4The Plantain Seed Polysaccharides can significantly improve thereparation of damaged liver cells, and then protect the normal function ofliver.
