Objective:To investigate the theraptic effect of berberine(BBR) to alcohol damaged heart of rats, the best dosage and its mechanism.Methods:Sixty specific pathogen free adult mate Sprague-Dawley (SD) rats were randomly divided into six group:(1) control group(10, distilled water),(2) small dose group(10, berberine100mg),(3)middle group(10, berberine200mg),(4) high group(10, berberine400mg),(5) trimetazidine group(10, trimetazidine20mg),(6) normal control group10, only distilled water). The rats of all five test groups were given alcohol56%for12weeks (5ml/kg/d the first week,10ml/Kg/d the second and third week,15ml/kg/d the fourth week). After12weeks, BW (body weight), HW (heart weight) and ultrasonic cardiogram were measured. The CRP, CK, CTnT, TG, TC, LDL-C, HDL-C were also measured. The myocardial pathological changes of myocardial cells were observed by HE stain. The apoptosis and expression of apoptosis-associated gene Bcl-2, Bax were measured by RT-PCR. The experimental data was statistically analyzed by independent-samples T test and mean difference between the two compares used q Test, there were significant when p<0.05.Results:The BW of rats of all five tested groups were lower than that of the normal control group, especially the middle and high dose group, as compared to that of the low dose group, trimetazidine group and control group (P<0.05). However, the HW were increased in the rats of control group and low dose group as compared to that of the other groups (P<0.05). The LVEDD, LVESD, LA of the rats of the control group and low dose group were increased than those of the middle, high dose group and trimetazidine group (P<0.05). Compare to that of middle and high dose group and trimetazidine group, the levels of CTnT, CK and CRP of the control and low dose group were increased significantly (P<0.05). The HDL-C levels of rats of all five tested groups were lower than that of the normal control group, The TG levels of rats of the low dose group, trimetazidine group and control group were higher than the other groups, however the levels of rats of TC, LDL-C of middle and high group were decreased (P<0.05).The myocardial fibers disarrayed and inflammatory cells in infiltrated in myocardial interstitial of the rats of the control and low dose group. However, only a few inflammatory cells infiltrated a small part of myocardial cells swollen and disarrayed in rats of the middle and high dose and trimetazidine group. Compared to that of the rats of normal control group, the expression of Bax of regulated significantly in rats of control and low dose group (P<0.05), Bcl-2/Bax decreased. However, the expression of Bax were only lightly elevated (P>0.05), Bcl-2/Bax elevated in the rats of middle, high dose group and trimetazidine group.Conclusion:The myocardial cells of the rats were damaged after long-term and excessive intake of alcohol. And it can be protected or lightened by taking certain dosage of BBR.
