The Protective Effect Of Berberine Preconditioning On Myocardial Ischemia Reperfusion Injury In Rats

Objective:To investigate the protective effect of Berberine preconditioning on myocardial ischemia reperfusion injury (MIRI) in rats and ignore its mechanism.Methods:60 SD rats were divided into 4 groups at random: Sham-operatedgroup,ischemia-reperfusiongroup,Berberine-pre treatmentgroup(0.1mg/kg/d),simvastatin-pretreatmentgroup(0. 1mg/kg/d).Rats of Berberine-pretreatment group and simvastatin-pretreatment group had been given Berberine and simvastatin respectively by via of peritoneal injection one time every day for seven days.At last time,all the rats had been given above-mentioned medicine by via of peritoneal injection 2 hours before cornonary artery had been ligated. While Rats of Sham-operated group and ischemia-reperfusion group had been given tales doses physiological saline.Then we ligated them at the middist and urpper 1/3 of anterior desending branch of rats left cornonary artery,1 hours later we cut the ligatore we occlused rat' s chest by via of hemostat,then gave them reperfusion for 2 hours,and we measured heartrate,LVSP, LVEDP,±dp/dtmax,after that we detected the SOD activity and MDA contents of the cardiac muscle,detected the blood lever of LDH and CK and measured infarct size by TTC staining. Meanwhile,we detected the rate of apoptosis deoxynucleotide by TUNEL,detected the the positive expression of Bax and Bcl-2 of ischemia-reperfusion of Rats cardiac muscle by immune of luorescence.Results:Compared with Sham-operated group,the model group of±dp/dtmax,the LVSP and the positive expression of Bcl-2 of Rats cardiac muscle and the SOD contents of the cardiac muscle of Berberine-treated group and simvastatin -treated group all decreased sharply.(p＜0.05 or p＜0.01),and the LVEDP and the blood lever of LDH and CK,the MDA contents of the cardiac muscle,Bax/Bcl-2 of Berberine -pretreatement group and simvastatin-treated group increased sharply(p＜0.01),the rate of apoptosis as well.(p＜0.01).Compared with ischemia reperfusion group,the model group of±dp/dtmax,the LVSP and the positive expression of Bcl-2 of Rats cardiac muscle and the SOD contents of the cardiac muscle of Berberine-treated group and simvastatin -treated group all increased sharply. (p＜0.05 or p＜0.01),and the LVEDP and the blood lever of LDH and CK,the MDA contents of the cardiac muscle,Bax/Bcl-2 of Berberine -pretreatement group and simvastatin -treated group decreased sharply(p＜0.01),the rate of apoptosis as well. (p＜0.01). Conclusions:Berberine preconditioning has protective effect on myocardial ischemia-reperfusion injury in Rats.The mechanism of its protective effect may relate to its effect of anti-oxidation and repression of apoptosis.
