Objective:to observe the influence about Xin kang granule impact on genetic expression of AVPmRNA of the hypothalamus、 AVPV2mRNA and AQP2mRNA of the kidney in chronic heart failure rats and to explore Xin kang granule in CHF the molecular biology mechanism of diuresis detumescence.Methods:SD rats were randomly divided into6groups:blank control group, model control group, Xinkang granule high、middle and low-dose groups, positive drug group.Intraperitoneal injection of adriamycin method(2mg/kg,1time weekly,in total10times) was adopted to establish the model of congestive heart failure. All SD rats were treated for6weeks,hen AVPmRNA、AVPV2mRNA and AQP2mRNA genetic expression were detected by means of RT-qPCR and collect24-hour urine (1times/d, continuous7d).Results:1. Contrast of urine of rat:Compared with normal group, the rest of the group of urine were higher(P<0.05),there were statistical signficance; Compared with model group, urine of the drug groups were iincreased(P<0.05), there were statistically significant (P<0.05); There was no statistically significant between Qili qiangxin capsule group and Xin kang granule group(P<0.05); There was litter difference between Xin kang granule groups(P>0.05), because of no statistical significance.2.Contrast of AVPmRNA、AVPV2mRNA and AQP2mRNA genetic expression:Compared with normal group, the rest of the group of AVPmRNA、AVPV2mRNA and AQP2mRNA were higher (P<0.05), there was statistical signficance; Compared with model group, AVPmRNA、AVPV2mRNA and AQP2mRNA of the drug groups were lower(P<0.05), there was statistically significant(P<0.05); There was no statistically significant betteen Qili qiangxin capsule group and Xin kang granule group(P<0.05); There was litter difference between Xin kang granule groups(P>0.05), because of no statistical significance.Conclusion:1. By inhibiting the hypothalamus AVP release,lower of AVPmRNA、AVPV2mRNA and AQP2mRNA genetic expression lead to water absorption decrease and increase of urine, which was probably one of its mechanism of action diuretic swelling of rat of chronic heart failure.
