Objective After generalizing the theories of employing traditional Chinese medicineexternal application therapy after fracture,evaluating the function and curative effectof the therapy in promoting fracture healing process,and based on the establishment ofrabbit fracture model, the thesis observes the change in broken bone edge and theformation of bone callus shadow after applying the Jiegu paste through medicalimaging; the fracture healing and the expression of BMP-2histologically andimmunohistochemically; also explores the acting mechanism of Tujia Jiegu paste inpromoting the fracture healing at the level of molecule.Method (1)Theoretical study:Through consulting substantial ancient and modernliterature, the thesis systematically organizes fracture the origin, etiology andpathogenesis, diagnosis and treatment; expounds the theoretical and practical value offracture application method.(2)Experimental study60adult healthy big-ear whiteJapanese rabbits are selected to build3mm bone defect of the left radius model.Randomly divided into the Jiegu paste experimental group and the model controlgroup,10rabbits in each group and every each week.1day after the model and theincision dried, the experimental group are applied Tujia Jiegu paste and bandage fixon the principle of small splint fixed treatment of fracture; the control group are notapplied but bandage fix with the identical splint. The experimental rabbits are killedwith air embolism method at the end of the2nd week,4th week and6th weekrespectively And then conduct bone specimen (paraffin embedded) biopsy, HEconventional staining,immunohistochemical detection, to explore the expression andchange of BMP-7in fracture healing at molecule level.(3)All the data are statisticallydealt with SPSS17.0software, said with x±s, and contrasted t-test with two samplemeans.Result (1)The theoretical discussion of the application method Application method,with a long history, convenient use, noticeable curative effect, economy and security,is an important part of external treatment of TCM and the common way to externally treat closed fracture.(2)Hematoxylin-eosin staining. Through light microscopicobservation, in the experimental group, there has considerable fibrous tissueconnection in the fracture area, which is earlier than in the control group,calcium saltgroup deposition, minor trabecular bone and cartilage composition in some area bythe2nd weekend. By the end of the4th week, fibrous callus formation in fractureends region, OB number and callus content in the experimental group are more thanthose in the control group.And lamellar bone formed, less cartilage existed, marrowcavity basically rechanneled in the experimental group. By the end of the6thweek,much more callus and lamellar bone formed,marrow cavity channeled andosseous connection time became earlier in the experimental group, visible trabecularbone and little cartilage composition lay in the control group.(3)Immunohistochemicaldetection At each time phase point, the measuredBMP-2MOD values in these twogroups are conducted sample mean t-test, showed remarkable difference(P<0.01); thevolume of BMP-72at each time phase point in the experimental group obviouslyhigher than that in the control group, and apparently prolonged the expressing time.Conclusion Fracture external application method, with a long history, convenient toapply is the common way to externally treat closed fracture. The application methodfor curing fracture with Tujia minority medicine Jiegu paste can improve the fractureblood circulation greatly, promote hematoma absorption and calcium deposition.Enhance and extend the volume of BMP-2at each time phase.
