Objective:To evaluate the effects of Anshenning syrup on improving sleep and explore its mechanism.Research Methods1、The research of Anshenning syrup affecting the hypnotic time on mice by pentobarbital sodium in threshold dosageExperimental animals were divided into six groups:blank control group,extazolam control group,ethanol control group,anshenning syrup high,medium and low dose.Each group had twenty.Each drug group were given extazolam (0.13×10-3g/kg),ethanol (1.6g/kg),anshenning syrup (0.96g/kg、0.48g/kg、0.24g/kg).Blank control group were given the same volume of distilled water.Administratered by gavage of seven consecutive days.Given pentobarbital sodium (50mg/kg) by intraperitoneal injection after the last administration thirty minutes. Record the rats’sleep latent period and sleep time.Results1、Anshenning syrup on the threshold dose of pentobarbital sodium in mice induced by sleep time effectsThe experimental results show that, Compared with the blank control group,Anshenning syrup high dose group and medium dose group could significantly prolong the sleep time (P<0.01, P<0.05), low dose group had a tendency to extend the sleep time,but there was no significant difference(P>0.05). Anshenning syrup high dose group could significantly shortern sleep latency (P<0.05), medium dose group and low dose group had a tendency to shorten sleep latency, but there was no significant difference (P>0.05). Compared with the ethanol control group, Anshenning syrup high dose group could significantly prolong the sleep time and shorten sleep latency (P<0.05), medium dose group had a tendency to prolong the sleep time and shorten sleep latency, but there was no significant difference (P>0.05). Compared with the extazolam control group, Anshenning syrup high dose group had a tendency to prolong the sleep time and shorten sleep latency, but there was no statistically significant (P>0.5).2、Anshenning syrup on under threshold dose of pentobarbital sodium in mice induced by sleep incidence:The experimental results show that, With the blank control group Anshenning syrup high dose group and medium dose group could significantly increase the incidence of sleep (P<0.01,P<0.05),low dose group had a tendency to increase the incidence of sleep,but there was no statistically significant(P>0.05).3^Anshenning syrup affecting sleep dose-effect relationship:The experimental results show that, Anshenning syrup on improving sleep effectappears certain dose-effect relationship, in a dose range, along with the increase of dosage, increase the efficacy, the optimal effective dose of0.96g/kg.4、Anshenning syrup affecting sleep to time-effect relationship:The experimental results show that, Anshenning syrup improving sleep effect appears certain time-effect relation ship, within the scope of certain time, along with the delivery day increased, increase the efficacy.Primarily prolong pentobarbital sodium in mice induced by sleep duration (P<0.01) and shorten their sleep latency (P<0.01).Effective delivery time is three days, the best delivery time for nine days.5、Anshenning syrup activitied mice sleep duration and sleep phaseThe experimental results show that, in the sleep time, compared with the blank control group, in the circadian24h,Anshenning syrup group mices sleep time very significantly prolonged (P<0.01),awaking time very significantly shortened (P<0.01). In the sleep phase, compared with the blank control group, Non-rapid eye movements sleep(NREM) and fast eye movement sleep (REM) extend in varying degrees, in which the extension of the role of Non-rapid eye movements sleep(NREM) is particularly significant (P<0.01). Compared with the blank control group, in the sleep time,the echol mices sleep time prolonged effect were not obvious(P>0.5), In the sleep phase, Non-rapid eye movements sleep(NREM) and fast eye movement sleep (REM) prolonged effect were not obvious(P>0.5).6、Anshenning syrup combined with the horse Xini (Flumazenil) of pentobarbital sodium in mice induced by sleep time effectsThe experimental results show that,in comparison with blank control group,given alone Anshenning syrup,were prolonged pentobarbital sodium in mice induced by sleep duration and shortened the latency(P<0.01); Anshenning syrup compared with Anshenning syrup+Flumazenilwere prolonged pentobarbital sodium(P<0.05) in mice induced by sleep duration and shortened the latency(P<0.05); fluorine horse Xini can significantly blocked diazepam and Cirenshen effective part and sodium pentobarbital synergy effect of.Conclusion1、Anshenning syrup has a role in improving sleep.2、The optimal effective dose of Anshenning syrup improve sleep is0.96g/kg. the best delivery time for nine days.3、In the circadian24h,Anshenning syrup can prolong total sleep time in freele moving mice. In the sleep phase, Anshenning syrup can prolong Non-rapid eye movements sleep(NREM) and fast eye movement sleep (REM), where the prolonged action of Non-rapid eye movements sleep(NREM) was particularly notable.4、The role of Anshenning syrup on improving sleep maybe through adjusting benzodiazepine receptor.
