Objective:To investigate the relative effect of synthetic borneol in mice sleep time and sleep phaseMethods:1. Based on the tracing and analysis of mice EEG for 3 consecutive days, and the feature research on sleep time and sleep phase in mice.Select 20 ICR male mice, under the conditions of anesthesia and cerebral stereotactic, buried cortex and EMG electrodes.Use 16 physiological signal recorder for 3 consecutive days tracings cortical electroencephalogram (EEG) and electromyography and analyzed in order to accumulate during the day and night time awakening, sleep time, and when the cumulative percentage of each sleep phase (Wake%, NREM%, REM%) is observed indicators, study changes in the mice sleep time and sleep phase.2. Based on the tracing and analysis of mice EEG for 3 consecutive days, research on effect of synthetic borneol on the sleep phase and the sleep time of mice.Select 40 ICR male mice, Randomly divided into blank group and Drug Administration Group,20 rats in each group. Under the condition of anesthesia and brain stereotactic, buried cortex and EMG electrodes, restore seven days, and 6:00 pm every day during recovery, administration group dosed 0.4g/kg intragastric administration of 7 days, the control group were given the same volume of saline. Based on the tracing and analysis of mice EEG for 3 consecutive days, and the feature research on sleep time and sleep phase in mice.1.1 Based on the tracing and analysis of mice EEG for 3 consecutive days the feature research on sleep time of mice.The experimental results show that:First day monitoring period:day 12 hours 20 mice average cumulative time of awakening 312.37min; cumulative average sleep time was 390.53 min; 20 mice 12 hours at night cumulative average awakening time was 397.16 min; cumulative average sleep time was 320.19 min.Second day monitoring period:day 12 hours 20 mice average cumulative time of awakening 312.30min; cumulative average sleep time was 386.11 min; night 12 hours 20 mice average cumulative time of awakening 407.31min; cumulative average sleep time was 307.33 min.Third days monitoring period:day 12 hours 20 mice average cumulative time of awakening 315.69min; cumulative average sleep time was 391.60 min; night 12 hours 20 mice average cumulative time of awakening 409.01min; cumulative average sleep time was 317.33 min.During the three days to monitor, cumulative time of 20 mice awakening day and night of continuous and cumulative sleep time no significant difference (P> 0.05)1.2 Based on the tracing and analysis of mice EEG for 3 consecutive days the feature research on sleep phase of mice.The results display that:During the first day of monitoring:day 12 hours 20 mice sleep phase Wake percentage average of 44.93%; the percentage of NREM sleep phase with an average of 49.26%; relative to the average percentage of REM sleep was 5.42%; phase 12 hours during the night of 20 mice sleep Wake percentage average of 55.99%; the percentage of NREM sleep phase with an average of 40.03%; percentage of REM sleep phase average of 4.74%.During the second day of monitoring:day 12 hours 20 mice sleep phase Wake percentage average of 45.68%; the percentage of NREM sleep phase with an average of 48.26%; relative to the average percentage of REM sleep was 5.69%; phase 12 hours during the night of 20 mice sleep Wake percentage average of 58.26%; the percentage of NREM sleep phase with an average of 38.40%; percentage of REM sleep phase with an average of 4.70%.During the third day of monitoring:day 12 hours 20 mice sleep phase Wake percentage average of 45.17%; the percentage of NREM sleep phase with an average of 48.72%; relative to the average percentage of REM sleep was 5.91%; phase 12 hours during the night of 20 mice sleep Wake percentage average of 58.70%; the percentage of NREM sleep phase with an average of 36.94%; percentage of REM sleep phase average of 4.69%.Monitoring for 3 consecutive days, day and night when sleep phase Wake, sleep phase NREM, the percentage of no significant difference (P> 0.05) with REM sleep.2. Based on the tracing and analysis of mice EEG for 3 consecutive days, and the feature research on sleep time and sleep phase in mice:Sleep time:during the day, the cumulative time of awakening 312.37min, cumulative sleep time was 390.53 min; night time was 397.16 min cumulative awakening cumulative sleep time 320.19min;Sleep phase:during the day, Wake, NREM, REM percentage was 45.26%, 47.56%,5.67%; night, Wake, NREM, REM percentage was 57.73%,39.12%, 4.71%. Tracings of mice during the phase when the sleep and sleep time was stable.2. Effects of Synthetic Borneol on sleep duration and sleep phases in mice for 3 consecutive days:During the day, the cumulative time of awakening were significantly shortened; cumulative sleep time were significantly prolonged; night, accumulation awakening time there was a significant prolongation:cumulative sleep time were significantly shortened.
