OBJECTIVE:The objective of this research was to notarize the optimal dose of Naojian Capsule by discussing it’s influence to neurological function damage degree of focal cerebral ischemia, cerebral infarction size, cerebral water content and the quality of rats. Also the impact of Naojian Capsule to the factor expression of FADD, caspase-8and caspase-3in Fas apoptotic pathway after focal cerebral ischemia is the research contents in order to probe the curative mechanism of this prescription against cerebral ischemia and the molecular mechanism of it’s Inhibition of neuron apoptosis.METHODS:1. SD rats was randomly equivalently divided into6groups, including Buyang Huangwu Decoction group,3Naojian Capsule groups (high dosage, medium dosage and low dosage), model group and sham operation group. Focal Middle Cerebral Artery Occlusion (MCAO) model of rats was copied by suture method in middle cerebral artery. Isopyknic sterile distilled water was gavaged in model group and sham operation group, other goups were in intragastric administration after molding.7days for a stage and1time per day. Neurological score was applied in the first day and seventh day and weight was recorded everyday. In the seventh day, we took the cerebral tissue to observe the TTC and the cerebral weight ratio of dry weight and wet weight.2. SD rats was randomly divided into4groups, including Buyang Huangwu Decoction group (n=50), Naojian Capsule group (n=50), model group (n=50) and sham operation group (n=10). Isopyknic sterile distilled water was gavaged in model group and normal group, other goups were in intragastric administration after molding. Blood sampling in aorta abdominalis and cerebral tissue acquirement were carried out in the first, third, seventh, forteenth and twenty-eighth days. Factor expression of FADD, caspase-8and caspase-3in cerebral tissue and blood were detected with immunohistochemical method and Elisa.RESULTS:1. All of the MCAO rats express obvious neurological function damage. There are no statistical significance in groups (p>0.05) except for sham operation group. Contrast to model group, the ratio of cerebral infarction size and cerebral water content percentage show significant reduce(p<0.05).2. Serological results show an obvious concentration increase of caspase-8and caspase-3in MCAO rats’serum. Serum level of Naojian Capsule group is statistically significant and postpone the peak of caspase-8and caspase-3contrast to model group.3. Immunohistochemical results suggest a certain number expression of FADD, caspase-8and caspase-3in all rats’ hippocampus CA1and cortex. Expression in Naojian Capsule group is statistically significant and show a significant decrease contrast to model group.CONCLUSION:1. The best dosage of Naojian Capsule is medium in7days after cerebral ischemia2. After the ischemia, there exist neurological defect, infarction in the brain, and encephaledema.3. Naojian Capsule has a function of reducing nerve cells apoptosis, protecting cerebral tissue and relieve the damage after cerebral ischemia by inhibiting expression of FADD, caspase-3and caspase-8.
