| Objective: To build the rat model of cerebral isehemia with intelligentimpairment,and explore neuroprotection mechanisms of shenlongjiannao capsulefor this model.Methods: Two-vessel occlusion were adopted to establish the cerebral ischaemiamodel.60rats were randomly divided into5groups: sham operation group, modelgroup, pisacetam group, shenlongjiannao capsule groups(low and high dosage).On the7d,14d and28d, the scores of neural function were observed. Afteradministrating intragastrically for28days, the learning and memory abilities weretested with the Morris water maze. The expression of ACh was detected withELISA in the brain tissue of rats; colorimetry was used to determine the contentsof Glu in brain tissue; H-E staining was used to observed the change of braintissue sections; immunohistochemistry was used to assay the expression of ChATã€NR1ã€BDNFã€TrkBã€CGRPã€NPY in hippocampus of cerebral ischaemia rats.Results:1The evaluation results for the rats of cerebral isehemia with intelligentimpairment showed that:compared with sham operation group,the neurologicalscores were significantly increased in model group(P<0.01); the escape latencywere significantly extended and the frequency of acrossing former platform weresignificantly increased in model group(P<0.01).2The results of neurological scores and Morris water maze test showedthat:compared with model group, neurological scores were significantly reducedin shenlongjiannao capsul group and pisacetam group(P<0.01, P<0.05);compared with model group, the escape latency and the frequency of acrossing formerplatform were significantly shortened in shenlongjiannao capsule group andpisacetam group(P<0.01, P<0.05).3The results of H-E staining showed that: compared with model group,thehierarchy of neurons was relatively clear,the number of Pyknosis and deeplystained cells were reduced,the numbers of denatured and necrosed neurocyte werelower in shenlongjiannao capsul group and pisacetam group.4The results of AChã€ChATã€Glu〠NR1ã€BDNFã€TrkBã€CGRPã€NPYdetermination showed that: the contents of ACh and the expression of ChAT weresignificantly increased in shenlongjiannao capsule groups and pisacetam groupcompared with model group(P<0.01, P<0.05); the contents of Glu and theexpression of NR1were significantly reduced in shenlongjiannao capsule groupsand pisacetam group compared with model group (P<0.01, P<0.05); theexpression of BDNF and TrkB were significantly increased in shenlongjiannaocapsule groups and pisacetam group compared with model group(P<0.01); theexpression of CGRP was increased and the expression of NPY was decreased inshenlongjiannao capsule group and pisacetam group compared with modelgroup(P<0.01, P<0.05).Conclusion: The shenlongjiannao capsule could improve the learning andmemory abilities in animal model of cerebral ischemia with intelligent impairment.Its mechanism may be related to reducing the necrosis of brain neurons, toenhance the effectiveness of cholinergic system, to regulating the release of Gluand the expression of NR1, against the excitotoxicity, to increasing the expressionof BDNF and TrkB, protect the damaged neurons,to regulating the expression ofCGRP and NPY to control the vasomotor state of cerebral. |
