Preliminary Study On The Pharmacodynamic Basis Of Naojian Capsule And Its Effect On Cerebral Blood Flow In Anesthetized Dogs

Objectives The study was to discussed the effect of Nao Jian Capsule on cerebral blood flow and cerebral vascular resistance in anesthetized dogs.Methods(1) NaoJian Capsule on studies of Preliminary study on the effective substance basis: In healthy adult dogs as the experimental object, dogs were randomly divided into blank control group(n = 2), Nao Jian Capsule group(4), Bu Yang Huan Wu Decoction group(n = 4). Continuous oral administration after 7 days, collecting the last administration after 1h anesthesia dogs medicated serum, plasma and cerebrospinal fluid(CSF), by HPLC?HPLC-MS/MS detection and analysis in serum, plasma and cerebrospinal fluid(CSF) of the active substance.(2) Effects of Nao Jian Capsule on cerebral blood flow in anesthetized dogs: in healthy adult dogs as experimental subjects randomly points for Nao Jian Capsule group, Bu Yang Huan Wu Decoction group, western medicine group and blank group, 5 rats in each group, transduodenal after drug administration were recorded and the comparison of different times of brain blood flow, cerebral vascular resistance(CVR), systolic blood pressure, diastolic blood pressure, heart rate changes.Result(1)Through in plasma, serum, cerebrospinal fluid(CSF) fingerprint analysis to determine the Naojian Capsule into the blood of the five components, mainly of Astragalus ononin, formononetin, Chuanxiong in Ligustrazine and senkyunolide A and Artemisia the lactone class; Bu Yang Huan Wu Decoction 7 into the blood components, mainly of linarin in Astragalus Coleus forskohlii isoflavone glycosides, formononetin, formononetin, Chuanxiong in Ligustrazine, ferulic acid, senkyunolide A and ligustilide class. Predictably Naojian Capsule and Bu Yang Huan Wu Decoction of blood components are similar, mainly from two flavor of Chinese medicine prescription Jun medicine Astragalus, adjuvant drug Ligusticum chuanxiong Hort.(2) By observing the experimental animals in each group at different time points of brain blood flow, brain vascular resistance, blood pressure and heart rate, and blank group compared to Naojian Capsule group anesthesia dog blood flow to the brain increases gradually, cerebral vascular resistance decreased gradually, the difference is statistically significant(P<0.05), compared with before administration there were also statistically significant(P<0.01), compared with before administration and Naojian Capsule in anesthetized dogs blood pressure and heart rate were not statistical significance(P>0.05). So we can know that the Naojian Capsule can obviously increase cerebral blood flow, reduce the cerebral vascular resistance.Conclusion The study from in vitro and in vivo efficacy and material basis of Naojian Capsule were preliminarily, basically to determined the Naojian Capsule main pharmacodynamic substance foundation mainly from and Bu Yang Huan Wu Decoction is some two kinds of traditional Chinese medicine that Radix Astragali, Rhizoma Chuanxiong, illustrates the material basis of brain healthy piece with and Yang also five soup consistent. At the same time through the observation of Naojian Capsule at different time points on anesthetized dogs of cerebral blood flow, cerebral vascular resistance, heart rate and blood pressure of confirmed cerebral Naojian Capsule can increase blood flow to the brain, reduce cerebral vascular resistance, thereby improving the brain blood circulation.
