| Objective:To explore the therapeutic effects and possible mechanisms of QinBojiajianfang to ulcerative colitis by detecting the expression of Aquaporin-4(AQP4) and Aquaporin-8(AQP8) in rats with ulcerative colitis.Methods:A total of48healthy male rats were randomly divided into four groups:the normal group, model group, OLZ group(western medicine group), QinBojiajianfang group(Chinese traditional medicine group),each group has12rats. In addition to the normal group,all of the rest three groups use an experimental rat model of ulcerative colitis which was established by giving trinitro-benzene-sulfonic acid(TNBS).Starting drug intervention after the model established successfully and the treatment will be continued for3weeks. Body weightã€nature of stool and mental state of the rats will be observed during the duration.At the end of treatment,taking colon tissues on all groups for CMDI score, and then getting some slice for HE dyeing for histological score and the other parts for detecting AQP4and AQP8using immunohistochemistry.Results:(1)The UC rats appear symptoms such as apathetic, lethargy, slump, lazy,,no appetite, soft stools, side walk, perianal pollution etc. The bloody mucopurulent stool and emaciation appeared after TNBS and ethanol are injected through the anus. The pathological findings of colon shown that mucous hyperemia, edema, plica reduced or even disappeared, ulcer formation, black and yellow of membrane attachment, or the formation of polyps, bowel thickening, bowel wall thinning and tissue adhesion.(2)The results detected by immune histochemical chemistry shown that the expression of AQP4and AQP8positive cells in the rats with syndrome of dampness stagnancy is less than in normal group, the number of positive cells and the average optical density decreased significantly, which have very remarkable difference compared with the normal control group (P<0.05). The expression of AQP4and AQP8in Traditional Chinese medicine group and Western medicine group increases significantly and shows a significant difference when compared with model group (P<0.05). There is a significant difference between Traditional Chinese medicine group and Western medicine group on the expression of AQP4(P<0.05).Conclusion:The experimental results show that:AQP4and AQP8in ulcerative colitis model rats were significantly higher expression; QinBojiajianfang with retention enema can effectively treat the rats with experimental ulcerative colitis, improves the general condition and alleviate the inflammatory damage of colon.These studies indicate that QinBojiajianfang can resist ulcerative colitis by the expression of AQP4and AQP8. |
