Effect Of Shaoyao Decoction On The Expression Of Aquaporins And Mucosal Permeability In Colonic Mucosa Of Rats With Internal Damp-Heat Type U

Objective: based on Shaoyao Decoction to explore the effect of Shaoyao Decoction on the expression of aquaporin in colon mucosa and the mechanism of Shaoyao Decoction in the permeability damage of intestinal mucosa in rats with damp-heat type UC by interfering with the expression of aquaporin.Methods: Establish the rat model of damp-heat UC,observe the general behavior,colon score,HE staining pathological section and detect the level of plasma GAS to evaluate the model of damp-heat UC.Use ELISA to detect plasma D-lactic acid and DAO levels,immunohistochemistry to detect the expression of AQP4 and AQP8 in colon tissue,and explore whether Shaoyao Decoction can repair the intestinal mucosal barrier function and improve the permeability of intestinal mucosa by interfering with the expression of mucosal aquaporinResults:(1)DAI score,colon tissue score and HE pathological score of rats in each treatment group were lower than those in the model group(P<0.01),which suggests that Shaoyao Decoction and mesalazine have therapeutic effect on UC rats with damp-heat accumulation;(2)After treatment,the blood GAS of rats increased(P<0.01),suggesting that Shaoyao Decoction can improve the immunological indexes of rat damp-heat model;(3)the expression of AQP4 and AQP8 protein in colon increased,suggesting that Shaoyao Decoction can improve the level of AQP4 and AQP8,and the effect of Shaoyao Decoction is significantly better than that of mesalazine(P<0.05);(4)After treatment,the level of D-lactic acid and DAO in blood of rats decreased(P<0.05),which suggests that Shaoyao Decoction can repair the barrier function of intestinal mucosa and improve the permeability of intestinal mucosa,and Shaoyao Decoction has advantages over mesalazine in improving blood D-lactic acid levels(P<0.01).Conclusion:(1)Shaoyao Decoction can improve the general condition and symptoms of UC rats with damp-heat accumulation,and reduce colon injury;(2)Shaoyao Decoction can increase the expression of AQP4 and AQP8 protein in colon tissue of rats;(3)Shaoyao Decoction can reduce the content of D-lactic acid and DAO in rat plasma;(4)Shaoyao Decoction may repair the intestinal barrier function and improve the permeability of intestinal mucosa by interfering with the expression of AQP4 and AQP8 aquaporins in colon tissue.
