| Purpose: In recent years, the research on podocyte of glomerular injury is increasinglymature.The podocyte is the main cell to maintain the structure and function of the glomerularbasement membrane, which participate in the function of glomerular filtration barrier.BecauseDesmin can be cytoskeleton reorganization when injury occurred in the podocyte, so we canconsider that Desmin is an expression of a specific damage of podocyte. The capsule of JieduTongluo Baoshen capsules by Professor Zheng Nan has a good effect in clinic.Method:10rats were selected for normal control group randomly in the80male Wistarrats,others were injected the STZ intraperitoneally.The model rats were randomly dividedinto:model group (DM), Jiedu Tongluo Baoshen capsule group,positive control group.Wetook rats urine and blood to measure blood and urine biochemical indexes after16weeksbefore all the rats were killed.Then weighed the body and kidney weight of rats. Pathologicalchanges were evaluated by HE stain in renal tissues; and the ultrastructures of Podocyte wereobserved by electron microscope，We use immunoturbidimetry assay to detect Desmin whichis a sign of rat models of podocyte proteins, and objective to evaluate the clinical efficacy andsafety of Jiedu Tongluo Baoshen capsules.Result: The FPG,blood fat,BUN,Scr,urine volume,urinary albumin and kidney weight ofmodel group were higher than that of normal control group, which of Jiedu Tongluo Baoshencapsule group and positive control group were lower than that of model group.The bodyweight of model group were significantly lower than that of normal control group,The bodyweight of Jiedu Tongluo Baoshen capsule group and positive control group were higher thanthat of model group.Compared with normal control group. Pathological changes wereevaluated by HE stain in renal tissues were mormal, and the ultrastructures of Podocyte wereobserved by electron microscope were not change，but that of model group (DM) weresignificantly changed,which of Jiedu Tongluo Baoshen capsule group and positive controlgroup were lower than that of model group.The expression of Desmin in model groupincreased markedly and the expression of Desmin in Jiedu Tongluo Baoshen capsules group decreased compared with model group.Conclusion: Jiedu Tongluo Baoshen capsule can markedly decrease the FPG,bloodfat,BUN,Scr,urine volume,urinary albumin,kidney weight of experimental diabetic rats,whichcan increase the body weight and lessen glomerular pathology and ultrastructre changes ofkidney, Jiedu Tongluo Baoshen capsule can lessen the expression of Desmin in renal tissue ofthe DM rats, and which plays an important role in preventing diabetic nephropathy from thestructural and functional damage of podocyte. |
