ObjectiveIn this study,the purpose of this study is to investigate the therapeutic effect of Xiaoshen Recipe on Diabetic kidney disease(DKD)by preparing Diabetic kidney disease rat model,giving Xiaoshen Recipe,a self-made traditional Chinese medicine,under the guidance of Jiedu Tongluo Yiqi Baoshen law,and to explore the mechanism of Xiaoshen Recipe on Diabetic kidney disease related inflammatory response by detecting the expression of inflammatory factors:TWEAK?MCP-1?IL-6?CRP.To reveal the effectiveness of the treatment of Diabetic kidney disease from the method of detoxification,Tongluo,yiqibaoshen and Xiaoshen Recipe,so as to provide theoretical basis for clinical treatment of Diabetic kidney disease.Method112 healthy male Wistar rats weighing 200-220 g.The Diabetic kidney disease animal model was made by injecting streptozotocin 60mg/kg into male Wistar rats 12 hours after fasting,and the blood glucose was measured 72 hours later.If the blood glucose was more than 16.7mmol/L,it was regarded as the success of the hyperglycemia rat model.Except for 12 rats in the blank group,all the rats successfully copied were divided into model group,losartan potassium group,high dose group of Xiaoshen Recipe and middle dose group of Xiaoshen Recipe according to the digital table 15 rats in the low dose group of Xiaoshen Recipe,1 cage for each 5 rats.Start the medication,measure the weight every week,and measure the blood sugar every two weeks.After 10 weeks of continuous gavage,24 hours urine was collected from all animals,and urine indexes were measured.Blood samples were collected from abdominal aorta under anesthesia to detect biochemical indexes.The effects of losartan potassium,high dose of Xiaoshen Recipe,medium dose of Xiaoshen Recipe and low dose of Xiaoshen Recipe on rats were evaluated.He and Masson staining were used to observe the renal pathological sections of Wistar rats,and the expressionof inflammatory factors such as tweek,MCP-1,IL-6 and CRP were determined by enzyme-linked immunosorbent assay.Then the mechanism of Xiaoshen Recipe,a traditional Chinese medicine,was discussed.Result1 Body weight,kidney weight and organ index:The kidney weight and organ index of DKD rats in the model group and the three groups of Xiaoshen Decoction group were increased compared with the blank group,and the body weight was reduced.The model group had extremely significant differences(P <0.001).These three indicators increased,there was a significant difference(P <0.01);losartan potassium and Xiaoshen formula in the middle and low dose groups of rats have statistical significance(P <0.05).2 Blood glucose: The GLU of DKD rats in the model group was significantly higher than that in the blank group,and there was a significant difference(P <0.01).The GSP value in the model group was also higher than that in the blank group,and there was a difference(P <0.05).In the Shenfang high and middle dose groups,the blood glucose decreased significantly compared with the blank group(P <0.05).The GSP values of the losartan potassium group and Xiaoshenfang high dose group were significantly lower than the model group.Significant difference(P <0.001);the small and medium dose group of Xiaoshen Recipe can reduce GSP to a certain extent,which has statistical significance(P <0.05).3 Blood lipid: TG in DKD rats in model group was significantly higher than that in blank group,which was statistically significant(P < 0.05).Compared with model group,TC in Xiaoshenfang high-dose group had the most significant decrease in TC,with significant differences(P < 0.01)The TG of Xiaoshen Fang decreased in high and medium doses and losartan potassium group,and there were differences(P<0.05).4 Urine protein and urine microprotein: The urine microprotein of the modelgroup and each treatment group were significantly higher than that of the blank group,and the UPro of the model group rats was significantly increased,which were extremely significant differences(P <0.001);UPro value of DKD rats in each group of Xiaoshen Decoction was lower than that in model group,which was statistically significant(P <0.05);urine microalbumin in each treatment group was significantly lower than that in model group,with extremely significant differences.Sex(P<0.001).5 Kidney function:Compared with the model group,the SCr,UCr,and BUN values of the losartan potassium group all decreased,which was statistically significant(P <0.05);the two indexes of UCr and BUN in the high-dose Xiaoshen formula group were significantly reduced There are extremely significant differences(P <0.001).The BUN and UCr of the DKD rats in the low and middle dose groups of Xiaoshen Recipe were significantly decreased,with extremely significant differences(P <0.001),while the SCr values were also reduced to a certain extent,with differences(P <0.05).6 Inflammatory factors:There were significant differences in MCP-1,IL-6 and CRP in the model group(P<0.001,P<0.01);tweek and MCP-1 in the high dose group of Xiaoshen Recipe were significantly lower than those in the model group(P<0.01);there was no significant difference in MCP-1 in the low dose group of losartan potassium and Xiaoshen Recipe(P>0.05);IL-6 and CRP in the high dose and middle dose groups of losartan potassium and Xiaoshen Recipe were significantly lower than those in the model group(P<0.05)There was significant difference(P<0.001),CRP value decreased significantly(P<0.001)and IL-6 in low dose group(P<0.05).ConclusionJiedu Tongluo Yiqi Baoshen method is an effective method for the treatment of DKD.Under the guidance of this method,Xiaoshen formula,a traditional Chinesemedicine,can increase the body weight of DKD rats,improve the state of rats,significantly reduce the blood sugar,blood lipid,urinary protein,play a good protective role in renal function,inhibit the development of inflammatory factors in serum,and delay the occurrence and development of DKD.1 The injection of streptozotocin 60 mg / kg is an ideal method for the preparation of diabetic nephropathy animal model.Its advantage lies in the occurrence of kidney problems caused by hyperglycemia,strong controllability,direct and accurate operation method,which well simulates the pathogenesis of diabetic nephropathy.2 Inflammatory injury plays an important role in the progression of diabetic nephropathy,and inflammatory factors such as TWEAK,McP-1,il-6 and CRP are involved in this process.Inflammatory cell infiltration and inflammatory factor increase are the cytological and molecular bases of "turbidities" in nephropathy.3 Jiedu Tongluo Yiqi Baoshen method is an effective method for the treatment of DKD.Under the guidance of this method,Xiaoshen formula,a traditional Chinese medicine,can increase the weight and kidney weight of DKD rats,significantly reduce the blood sugar,blood lipid,urine protein,and play a good role in protecting the renal function.4 The mechanism of XiaoShen Recipe in the treatment of diabetic nephropathy animal model may be that it can inhibit the inflammatory development of DKD rats by reducing the expression level of inflammatory factors such as tweek,MCP-1,IL-6and CRP,and reduce renal damage,which is more obvious than losartan potassium alone.
