Objective:In this study,to investigate the regulatory effect of electroacupuncture(EA) atWeishu(Back-Shu acupoint of stomach) and Zhongwan(Front-Mu acupoint of stomach)points on the inteagastric pressure in rats with gastric distension and its neuralmechanism as a breakthrough point,Through the influence of electroacupuncture atBack-shu and Front-Mu points on the inteagastric pressure in rats on gastric internalpressure,levels of Cholecystokinin-8、Gastrin of serum,the expression of c-fos inDorsal vagal complex. Exploring the regulatory effect of electroacupuncture atBack-shu and Front-Mu points on the inteagastric pressure in rats with gastricdistension,verifying the effect of central targets and patnways of cooperativity ofcombination of electroacupuncture at Back-shu and Front-Mu points on gastricmotility,partily to reveal the neurobiological basis of Back-shu and Front-Mu pointstheory,and to guide clinical improve the level of diagnosis and treatment.Methods:Healthy adult male SD rats were randomly divided into several groups. The intragastricpressure and gastric motility in rats were recorded by homemade balloon.; Method of cuttingthrough the lesion and diaphragm of DVC region of the rat vagal intervention,to observe the effectof electroacupuncture at Back-shu and Front-Mu points on the inteagastric pressure in rats withgastric distension;The levels of CCK-8and gastrin in serum of rats in each group were assayed byradioimmunoassay. The expression levels of c-fos in dorsal vagal complex were assayed byimmunohistochemistry.Results:1. Changes of the intragastric pressure:(1)Compared with the acupuncture,theamplitudes of intragastric pressure were increased significantly in the three EA treated groups (p<0.01); and they were statistically increased in Zhongwan plus Weishu groupand Weishu group compared with those in Zhongwan group (p<0.05or p<0.01); EAhad no significant effects on the frequency of gastric movement(p>0.05)(.2)Comparedwith model group,the amplitudes of intragastric pressure were reduced significantly inDVC damaged group and damage plus EA group (p<0.05); The vagotomy group andvagotomy plus EA group significantly decreased amplitude intragastric pressure (p<0.05);In comparision with those in the Sham lesion group,the amplitudes of intragastricpressure were reduced significantly in the DVC damaged group and damage plus EAgroup (p<0.05); and they were statistically reduced in the vagotomy group and vagotomyplus EA group compared with those in sham operation group (p<0.05).2. Changes of serum CCK-8and gastrin: Compared with those in the modelgroup,the serum levels of CCK-8were significantly reduced in Zhongwan group,andZhongwan plus Weishu group (p<0.05or p<0.01),and the serum levels of gastrin weresignificantly increased in,Zhongwan group and Weishu group (p<0.01).3. Changes of the expression of c-fos in dorsal vagal complex:(1)Incomparision with model group, the expressions of c-fos in dorsal vagal complex wereincreased significantly in the each EA groups (p<0.01); and they were statisticallyincreased in Zhongwan plus Weishu group compared with those in Zhongwan group,and Weishu group (p<0.01); EA had no significant effects on the expressions of c-fos(p>0.05).Conclusion:1. EA at “Weishu”,and “Zhongwan”,especially EA at “Weishu” combinedwith “Zhongwan”,can significantly strengthen rat intragastric pressure,the changes ofthe levels of CCK-8and gastrin in serum may be coused by the combination ofback-Shu and front-Mu acupoints regulating gastric movement.2. EA at Back-shu and Front-mu points of Stomach and gasreic distentioninformation in the medulla oblongata DVC aggregation and integration,indicating that DVC paticipate in regulating gastric movement with combination of back-Shu andfront-Mu acupoints.Explaining the central control target of combination of back-Shuand front-Mu acupoints3. Electroacupuncture at Back-shu and Front-Mu point to regulated the stomach movementdepends on the vagus nerve integrity. Effect of electroacupuncture on gastric Back-shu andFront-Mu points the regulation of gastric motility may be achieved through DVC-vagal gastricpathway.
