ObjectiveIn this study, we use the Double Dark Xuanbi Prescription, a Traditional Chinese medicine in the treatment of rheumatoid arthritis, as the research object. Safety of delivery of this drug is investigated through the acute toxic reaction in mice and pesticide effect of it is investigated through anti-inflamatory and analgesia experiments on mice. Improved prescription was made by adding Rhizoma Anemarrhenae and Radix Paeoniae Alba into the original prescription after consulting the literature to make a comparison of pesticide and mechanism in the treatment of rheumatoid arthritis between original prescription and improved prescription through anti-inflamatory and analgesia experiments on mice and the curative effect and immunoregulation experiment on adjuvant arthritis of rats.Methods1. Experiment of acute toxic on the whole mixed prescription is done to testify its safety.2. Toxic substance is found out according to the original broken prescription acute toxic experiment on the components of the fixed prescription.3. Make a series of pharmacodynamic research (anti-inflamatory effect、analgesia effect) on the whole prescription to testify its pesticide effect.4. Two kinds of herbs are added into the original prescription as the improved prescription after consulting the literature and then do a series of pharmacodynamic experiments (anti-inflamatory effect、analgesia effect、immunological effect、histopathologic effect) to investigate the pesticide effect and its therapeutic mechanism of both prescriptions preliminarily.Results1. Maximum tolerance limit of gavage on mice of the fixed prescription is4.3g/kg which is equivalent to32times the clinical adult dosage, this indicate that the fixed prescription is safe and low toxicity.2. The original broken prescription experiment indicates that its toxicity mainly comes from Betel nut and Artemisia annua 3. The anti-inflamatory and analgesia experiments of the fixed prescription indicate that it has significant anti-inflamatory and analgesia effect.4. Pharmacodynamic experiments on improved prescription and original prescription indicate that anti-inflamatory and analgesia effect of the improved prescription is better than that of the original prescription, but there is no significant difference between them, both treatment effect on rheumatoid arthritis of rats are obvious. The curative effect of the improved prescription is better than the original prescription. Inhibiting the inflammatory factors (IL-6、TNF-α) rather than promoting suppression of inflammatory factors is its mechanism of the treatment of rheumatoid arthritis.ConclusionDouble Dark Xuanbi Prescription,which has a good effect on anti-rheumatoid arthritis, has a very good market development potentiality. Pesticide effect of the treatment of rheumatoid arthritis of rats of the improved Double Dark Xuanbi Prescription is of priority. Maybe its mechanism is that it inhibits the inflammatory factors (IL-6、TNF-α) rather than promoting suppression of inflammatory factors...
