| Objective： Detection the expressions of TGF-β1, HIF-1α and VEGF in gastric cancertissues and corresponding adjacent noncancerous tissues and investigate their changesin expression in gastric carcinoma, analysis the relationship between their expressionsand clinic pathological parameters in gastric cancer.Methods: RT-PCR was used to detect the expressions of TGF-β1, HIF-1α, VEGFmRNA in gastric carcinoma specimens and corresponding adjacent normal tissue.Immunohistochemistry was used to detect the expressions of TGF-β1, HIF-1α, VEGFProtein in gastric carcinoma specimens and corresponding adjacent normal tissue.Analysis the relationship between TGF-β1, HIF-1α and VEGF expression with gastriccancer patient’s histological type of tumor, metastasis and TNM staging etc. Explorethe possibility of using these three proteins as the prognosis of patients with gastriccancer indicator.Results： In gastric carcinoma TGF-β1, HIF-1α and VEGF positive rates were62.7%、59.3%and63.3%; in adjacent tissues, the positive rates were10.7%,22%and13.3%.Tumor tissues and adjacent tissues there were significant differences (P <0.05),between tumor tiusserPatients and adjacent tissues. TGF-β1positive expressionshowed higher incidence of lymph node metastasis (P <0.05) and distant metastasis (P<0.05) than those with TGF-β1negative expression. The expressions of HIF-1α andVEGF were significantly correlated TNM stages, metastasis and Depth of invasion ingastric carcinoma (P <0.05). In the experiment we analyzed expressions of TGF-β1 and VEGF, HIF-1α, results showed that expression of VEGF and TGF-β1expressionwas significantly correlated (P<0.05); the expressions of HIF-1α and VEGF havesignificant difference, with statistical significance (P<0.05)Conclusions: The expressions of TGF-β1, HIF-1αand VEGF were closely related tothe biological characteristics in gastric carcinoma; Detection of TGF-β1, HIF-1α andVEGF may have certain value in predicting tumor prognosis invasion and metastasis. |
