The Expression And Clinical Significance Of VEGF And SDF-1α In Gastric Carcinoma Patients' Serum

Objective:To investigate Serum levels of vscular endothelial growth factor(VEGF) and stromal cell derived factor-1α(SDF-1α) in the pre-operation and post-operation patients with gastric carcinoma and explore its role in the development of gastric carcinoma;Through analyze the relationship between of them,illuminate the possible mechanism of action between them.Methods:ELISA quantitative analysis was used to examine the serum levels of VEGF and SDF-1αin thirty-five cases of gastric carcinoma and seventeen healthy controls.The relationship between VEGF and SDF-1αand clinicpathological factors were statistically analyzed.Results:As is shown,in the pre-operation group,VEGF and SDF-1αlevels in serum of the patients with gastric carcinoma were higher than that of the normal people(P＜0.01);but VEGF and SDF-1αlevels decreased obviously after operation(P＜0.01).In addition,the serum VEGF levels were related to the size, lymphatic metastasis and clinical stage of tumor(P＜0.05) but not associated with patient's age,sex and the degree of tumor differentiation(P＞0.05);the serum SDF-1αlevels were related to the lymphatic metastasis,clinical stage and local infiltration depth of tumor(P＜0.05) but not associated with patient's age,sex, size and the degree of tumor differentiation(P＞0.05),moreover,the SDF-1αlevels in serum of patients with gastric carcinoma were significantly related to the VEGF levels(pre-operation r=0.470,P＜0.01;post-operation r=0.510, P＜0.01).Conclusion:The level of VEGF and SDF-1αin serum of the gastric carcinoma were significantly higher than healthy controls.On the one hand, VEGF is play an important role in the development,infiltration and metastasis of tumor.Besides the function of stimulating angiogenesis by paracrine mechanism,VEGF possibly use autocrine signaling pathway interact with SDF-1α/CXCR4 biology axis,and mediate the infiltration and metastasis of gastric carcinoma.On the other hand,SDF-1αhas chemotaxis to tumor cells and it is an important factor in the metastasis of gastric carcinoma.
