| Purpose: Research on compound JianGu granules for the treatment of glucocorticoid‐induced avascular necrosis.Method: This test uses ethanol refluxing extraction, with characteristics of the content of total flavonoids as investigation index, make L9(34) orthogonal experiment design; Clean level after establishing experimental rabbit model of glucocorticoid‐induced avascular necrosis were given0.13,0.065,0.033g/mL of compound particles extract and physiological saline intervention, and not build mold blank control group. BMD decrease experimental rabbit model control group, the femoral osteoporosis, bone trabecula and easy to slice, triglycerides and total cholesterol were significantly increased, empty pit rate high, intramedullary fat cells increased, high, mediumand low dose group and drug situation by reduce the pathological changes, least change at low dose group. Granulating process: drug extract after decompression enrichment, dry grinding to powder, dextrin to do excipient,90%ethanol do wetting agents, drugs and material ratio of1:1.5.Conclusion: According to the properties of the active ingredients in medicines, extracted by ethanol reflux method alcohol soluble active ingredients of drugs, we adopted orthogonal design method, the ethanol concentration, ethanol ratio, extraction time and extraction times conditions screening, ultimately determine the optimal extraction process.Compound JianGu particles can improve glucocorticoid‐induced avascular necrosis change of triglycerides and total cholesterol in the blood, improving fat metabolism disorder of bone marrow cells; Also can promote osteoblast proliferation and differentiation in vitro maturation, inhibition of osteoclast activity, promote the femoral bone regeneration, the changing environment is change for the better in the femoral head, osteoporosis, prevention and control of hormones cause for compound JianGu particle therapy offers a preliminary theoretical basis hormone osteonecrosis. |
