Objective:Observing the intervention that adenylate cyclase agonist Forskolin acting on amniotic epithelial cells(WISH cells)to the Aquaporin8(AQP8) and Aquaporin9(AQP9).Methods:Divided amniotic epithelial cells(WISH cells) into different concentration of Forskolin(0μmol/L,1μmol/L,10μmol/L,50μmol/L,100μmol/L,200μmol/L),24hours in vitro culture,using real time RT-PCR and Western blot to detect the mRNA and protein expression of AQP8、AQP9.Results:The results shows that the expression of AQP8、AQP9mRNA and protein in the control group(0μmol/L) and1μmol/L group have no difference(P>0.05),means that the effect of1μmol/L Forskolin is not obvious; The rest of the various concentration group compared with control group, have significant differences(P<0.05), and it is on the rise along with the concentration gradient, reaches its peak in100μmol/L, when200μmol/L drop, but still higher than the control group.Conclusion:Forskolin,the adenylate cyclase agonist have raised effect of AQP8、AQP9mRNA and protein in amniotic epithelial cells(WISH cells), and has a dose dependent relationship. |
